摘  要:目的:观察游泳运动干预后肥胖大鼠血清及下丘脑Ghrelin水平的变化,探讨其在肥胖及代谢紊乱中的作用。方法:SD大鼠50只随机分为普食(NC,n=10)组和高脂(HD,n=40)组。筛选出成功建立肥胖模型20只随机分为肥胖对照(OB)组、游泳(SWI)组,各10只。SWI组予以每天1 h,每周5 d,共行6周游泳训练;OB组及NC组予等剂量蒸馏水灌胃。分别测定干预6周前后各组体重(BW)、空腹血糖(FPG)、三酰甘油(TG)、总胆固醇(TCH)、空腹胰岛素(FINS)、胰岛素抵抗指数(HOMA-IR)及血清Ghrelin。免疫组织化学法检测各组大鼠下丘脑Ghrelin表达水平。结果:干预前,OB组、SWI组均较NC组BW、FPG、TG、TCH、FINS、HOMA-IR显著升高,Ghrelin明显降低(P<0.01);干预后,SWI组均较OB组BW、TG、TCH、FPG、FINS、HOMA-IR明显降低,Ghrelin水平升高(P<0.01)。免疫组织化学显示OB组下丘脑Ghrelin表达较NC组降低(P<0.01),SWI组较OB组升高(P<0.05)。结论:游泳运动可改善胰岛素敏感性并减轻体重,降低肥胖大鼠Ghrelin水平。Objective:To observe the changes of Ghrelin level in serum and hypothalamus of high-diet induced obesity rats after 6-week swimming exercise, and to investigate the association of Ghrelin with obesity and metabolic syndrome. Methods : Fifty SD rats were randomly divided into normal diet group( NC,n = 10) and high fat diet group( HD,n = 40). Obesity models (n = 20) were established from the HD group, and equally assigned to obesity group( OB ) and swimming exercise group(SWI). The SWI group received standard swimming exercise 1 hour/day,5 day/week for 6 weeks, and the other two groups were given distilled water. Their body weight( BW ) , fasting plasma glucose ( FPG ), triglyceride ( TG ), total cholesterol ( TCH ), fasting insulin ( FINS ), HOMA index and fasting Gbrelin were measured 6 weeks before and after the intervene;the Ghrelin expression in the hypothalamus was detected by imnmnohistochemistry. Results:The BW,FPG,TG,TCH,FINS levels and HOMA-IR in the OB group and in the SWI group were increased and the Gbrelio level was decreased in contrast to the NC group before the intervene (P 〈 0.01 ) ;the BW, FPG, TG,TCH, FINS levels and HOMA-IR in the SWI group were decreased and the Ghrelin level was increased compared to that in the OB group(P 〈 0. 01 ) after tile intervene. Immunohistochemistry results showed that the Ghrelin expression level was increased in the hypothalamus of the OB group but was down-regulated in the SWI group( P 〈 0.05 ). Conclusions:Swimming exercise can improve the insulin resistance and reduce the weight and Ghrelin levels in obesity rats.
