卵黄抗体代替血清抗体评价种鸡群病原感染状态的研究

摘  要:为了解卵黄抗体与血清抗体间的相关性,分别采集一定数量的莱芜黑鸡、海兰褐蛋种鸡和SPF白来航鸡的血清、鸡蛋,所有鸡蛋与血清样品均一一对应。用禽网状内皮组织增生症病毒(R正v)、呼肠孤病毒(REOV)、I亚群禽白血病病毒(ALV—J)、AB亚群禽白血病病毒(ALV-AB)、禽传染性贫血病毒(CAV)以及传染性法氏囊病病毒(IBDV)等6种ELISA检测试剂盒,分别检测血清与卵黄中的抗体水平,用SPSSStatistics17.0程序比较分析血清与卵黄抗体OD值的相关系数,从而获得针对ELISA检测卵黄抗体的最佳稀释倍数。结果显示,23-38份卵黄样品中REV、REOV、ALV-J、ALV—AB、CAV以及IBDv的抗体检测最佳稀释比例分别为1:200、1:400、1:250、1:200、1:20和1:500,与血清中抗体OD值的相关系数分别为0.982、0.939、0.927、0.993、0.935和0.989;检测结果还表明,最佳稀释度卵黄与血清样品的阴阳性符合率为96%-100%。本研究显示,卵黄可代替血清样品用ELISA检测试剂盒检测,以判断种鸡群的病原感染状态。In order to understand the correlation between egg yolk antibody and serum antibody, the serum and eggs from Laiwu black ehicken, Hy-Line Brown egg-laying chicks and SPF White Leghorns were collected,whose number of samples were corresponding. The antibody level of REV,REOV,ALV-J,ALV-AB,CAV and IBDV in the serum and egg yolks was detected with the 6 ELISA test kits,respectively. And then the optimization dilution multiple of egg yolk antibody was acquired after analyzing the correlation coefficients of OD value between egg yolk antibody and serum with the SPSS Statistics I7.0 program. The correlation analysis showed that the optimization dilution multiples of REV, REOV, ALV-J, ALV-AB, CAV and IBDV antibody in 23 to 38 egg yolk samples were 1:200, 1:400,1:250, 1:200,1 : 20 and 1 : 500,while the correlation coefficients of OD value were 0.982,0.939,0.927,0.993,0.935 and 0.989,respectively. In addition,the coineidence rate of positive or negative results between egg yolk samples in the optimization dilution multiple and the serum samples was 96% to 100%. It suggested that eggs yolk could be used with ELISA test kits to reflect the state of pathogenic infection instead of serum.
