柴胡加龙骨牡蛎汤抗抑郁症剂量配比与量效学研究

摘　　要：目的 :探讨柴胡加龙骨牡蛎汤抗抑郁症的剂量配比与量效学关系。方法 :以小鼠悬尾不动时间、5-羟色胺酸诱导小鼠甩头次数为指标,将柴胡加龙骨牡蛎汤拆方为单味柴胡、小柴胡类方组(去柴胡)、非小柴胡组(桂枝、茯苓、大黄、龙骨、牡蛎),用均匀设计实验方法对柴胡加龙骨牡蛎汤进行拆方研究。结果 :柴胡组、非小柴胡组能显著缩短小鼠悬尾不动时间,增加5-羟色胺酸诱导小鼠甩头次数。同时在柴胡组剂量水平取2.5 g/kg、小柴胡组4.5g/kg、非小柴胡组5 g/kg,全方柴胡加龙骨牡蛎汤的抗抑郁作用最强。结论 :均匀设计是研究中医古方一个简单而有用的工具,它既能找出处方中药物的最佳配比,又不需要拆方研究即能获得处方中每一味药物的作用性质,更能体现研究中医处方的整体观。Objective： To explore the ratio-dosage and dose-effect of Chaihu Plus Longgu Mull Decoction （ CD ） on antidepressant. Methods ： The antisant effect of CD was evaluated with forced swimming test and 5-HTP-induced head-twitches model in mice. The CD was splited single Chaihu （ SX ）, Xiaochaihu Decoction （ XD ）, NON Xiaochaihu Decoction （ NXD ）, using the uniform design experiment method for CD opening party research. Results ： XD and NXD can remarkably shorten mice forced swimming time, increase 5-HTP-induced head-twitches times. At the same time SX dose of 2.5 g/kg, XD dose of 4.5 g/kg, NXD dose of 5 g/kg, the whole antidepressant action was the strongest. Conclusion： Uniform design is a simple and useful tool to study CM prescriptions. It can find out the optimal proportion of the prescription drugs and obtain each drug＇s property without aparting the prescription, greatly reflecting the overall concept of Chinese medicine prescription.
