摘  要:以中花2号无菌苗为材料,对愈伤组织诱导和细胞悬浮培养的最佳条件进行探讨,并对悬浮细胞的生长特性进行检测,建立了花生悬浮细胞培养体系。结果表明,叶片是诱导愈伤组织和进行悬浮培养的理想外植体,愈伤组织最佳诱导培养基为MS+6-BA 2.0mg/L+2,4-D 2.0mg/L,悬浮培养适宜培养基为MS+6-BA 2.0mg/L+2,4-D 2.0mg/L+KT 1.5mg/L。悬浮细胞生长呈"S"型曲线,培养12d达到最大生长量,最适继代周期为12d左右,在悬浮培养过程中,细胞数量呈现先上升后下降的趋势,培养液的pH和电导率变化则均为先下降再回升最后趋于稳定。With Zhonghua2 as material, the optimum culture conditions of callus induction and cell suspension culture were studied, the growth characteristics of suspension cell were detected, and a cell suspension culture system was established. The results showed that leaf was the best explant for callus induction and cell suspension culture, the optimal medium for callus induction was MS-+- 6-BA 2.0 mg/L+2,4-D 2.0 mg/L, the optimal formula for suspension culture was MS+6-BA 2.0 rag/L+ 2,4-D 2. 0 mg/L + KT 1. 5 mg/L. The growth of suspension cells exhibited an S-shaped curve, growth mass reached the maximum after 12d culture, the optimal subculture cycle was around 12d. In the process of suspension culture, the suspension cell number exhibited the change trend: firstly increased, then decreased. The change trends of pH value and conductivity showed the same: firstly decreased, then increased, and then became stable.
