摘  要:人绒毛膜促性腺激素β亚基 (h CGβ)羧基端 1 0 9~ 1 45残基组成的 3 7肽 (h CGβ-CTP3 7)是 h CG分子所特有的片段 ,并存在 h CG的特异表位 (抗原决定簇 )。为提高h CGβ-CTP3 7肽的免疫原性 ,本研究构建 h CGβ-CTP3 7四聚体 c DNA,克隆入载体p QE60 ,获得含 (h CGβ-CTP3 7) 4 c DNA的表达质粒 p QE60 -(h CGβ-CTP3 7) 4 ,在大肠杆菌X-Blue中进行表达 ,得到 h CGβ-CTP3 7四聚体。Western blotting结果表明 ,该四聚体分子量约为 2 0 k Da,并能与抗 h CG抗体结合 ,且结合能力比 h CGβ-CTP3 7二聚体强。因此 ,我们设想 h CGβ-CTP3WT5BZ] To enhance the immunogenicity of the carboxyl terminal peptide of β hCG, which consists of 37 amino acid residues (109~145) and exists the specific epitope (antigenic determinant) of hCG, hCGβ CTP37 tetraploid cDNA was constructed to string four hCGβ CTP37 cDNAs together. Then, hCGβ CTP37 tetraploid cDNA was subcloned into the E.Coli expression vector pQE60 to construct the expression vector pQE60 (hCGβ CTP37) 4, and pQE60 (hCGβ CTP37) 4 was subsequently transferred into E.coli X Blue to express the recombinant (hCGβ CTP37) 4. The result of Western blotting showed that the tetraploid of hCGβ CTP37 had apparent molecular weight of 20 kD a and had the comparatively stronger anti hCG antibody binding activity than the diploid of hCGβ CTP37. Therefore, we assume that the tetraploid of hCGβ CTP37 may have potential application as the immunogen of anti hCG fertility regulatory vaccine.
