摘 要:目的 探索α2 受体拮抗剂咪唑克生能否增强 5 HT1A受体激动剂丁螺环酮的抗冲突作用。方法 采用焦虑动物模型Vogel冲突饮水实验。 结果 低于有效剂量的咪唑克生 (0 1~ 1 0mg·kg-1)当与低于有效剂量的丁螺环酮 (0 3mg·kg-1)合用后 ,以剂量依赖性的方式 ,明显增加丁螺环酮的冲突饮水次数 ,当与有效剂量的丁螺环酮 (1 0mg·kg-1)合用后 ,抗冲突饮水次数的改变率从单用时的 +10 9%增至合用时的 +140 %~ +5 85 %。同时 ,合用后对大鼠自发性饮水没有影响。结论 咪唑克生能增强丁螺环酮的抗冲突作用 ,提示去甲肾上腺能和 5 羟色胺能系统之间可能存在交互作用 ,共同调节冲突行为。AIM To explore whether the α 2 adrenoceptor antagonist idazoxan can potentiate the anticonflict action of 5 HT 1A receptor agonist buspirone. METHODS The Vogels drinking conflict test was used. RESULTS Buspirone, at the dose which had no action (0 3 mg·kg -1 ), significantly produced dose independent increases in punished drinking when combined with suboptimal doses of idazoxan (0 03~1 0 mg·kg -1 ) Similarly, combinations of an effective dose of buspirone (1 mg·kg -1 ) and suboptimal doses of idazoxan (0 1~1 0 mg·kg -1 ) markedly enhanced the increases in the rate of punished responding elicited by buspirone (1 mg·kg -1 ) when adminitered alone (alone: +109%; in combinations: +140%~+585%). Meanwhile, buspirone when co administering idazoxan did not affect the number of licks during the unpunished situation as compared to controls. CONCLUSION Idazoxan can potentiate the anticonflict actions of buspirone and suggest a possible interaction between the noradrenergic and serotonergic systems in the mediation of the conflict behavior.
