摘  要:为了研究DNA甲基化对草莓休眠的影响,进一步阐明草莓休眠的机制,在温度5℃、光照8h处理条件下对全明星和达赛莱克特2个草莓品种进行休眠解除,研究在此过程中总DNA甲基化水平及相关基因表达变化。结果表明,在解除草莓休眠过程中,2个草莓品种新展开叶片中总DNA甲基化水平均降低。甲基转移酶FaDRM(domain-rearranged methyltransferase)基因在2个草莓品种中保持较低表达水平。转葡萄糖基酶FaROS1(repressor of silencing 1)基因在全明星和达赛莱克特中分别从第4周和第3周开始显著上调表达,FaROS1基因的上调表达可能是引起DNA甲基化水平降低的原因之一。The objective of this work was to clarify the effects of DNA methylation on dormancy release and simultaneously to learn the mechanisms of strawberry dormancy. In this study, different levels of genomic DNA methylation within young leaves were identified to investigate their effects on strawberry dormancy release and at the same time, the related genes expression levels were checked under controlled temperature and photoperiod conditions. Our experimental results clearly indicated that, when dormancy was released under 5 ~C, 8 h, total DNA methylation decreased in both cultivars. The FaDRM expression was maintained at low level throughout the process in both cultivars. The expression pattern of FaROS1 in All Star began to increase significantly after 4 weeks and in Darselect to increase significantly after 3 weeks. The up-regulation of FaROS1 may contribute to the increased total DNA methylation level.
