摘　　要：以4-氯乙酰乙酸乙酯为原料,首先与丙醛缩合后,由溴化钠进行氯原子交换制得溴化物,再与硫脲成环,碳酸二叔丁酯保护氨基,最后乙酯水解制得(Z)-2-(2-叔丁氧羰氨基噻唑-4-基)-2-戊烯酸目标产物,收率达到76%,纯度达到98%。该方法简单易行,收率高,纯度好,是一条适合工业化生产的新合成方法。(Z)-2-(2-tert-butoxycarbonylzole-4-yl)-2-pentenoic acid is a key intermediate for preparing cefcapene pivoxil as the fourth generation Cephalosporin antibiotics.Herein,(Z)-2-(2-tert-butoxycarbonylzole-4-yl)-2-pentenoic acid was prepared by the reaction of 4-chloroaceto-acetic ester with propanal,substitution of the chlorine by sodium bromide,closed ring reaction with thiourea,and protection of amino-group by the(BOC) 2 O,and hydrolysis of ester with the yield of 76%.This new synthetic method has the advantage of simpler operating procedure,high yield,good purity and applies to industrialization easily.
