摘 要:目的:观察大鼠脊髓后角浅层内δ阿片受体是否存在于初级传入C纤维内,为脑啡肽在痛觉初级传入调制中的突触前抑制机制提供更为可靠的形态学依据。材料与方法:应用双标(免疫)组织化学电镜技术标记大鼠脊髓后角浅层(Ⅰ~Ⅲ层)凝集素Ⅰ-B4结合位点和δ阿片受体。结果:大量轴突及轴突终末呈δ阿片受体阳性免疫反应,特别是在双标切片上,观察到约40%的凝集素Ⅰ-B4结合位点阳性纤维及其终未内也含有δ阿片受体。结论:脊髓后角浅层内的δ阿片受体部分存在于传递伤害性信息的初级传入C纤维中,以介导阿片物质脑啡肽对初级传入C纤维的突触前抑制作用。Objective: To provide morphological evidence for presynaptic inhibition mechanism of enkephalin in modulating primary input of pain by observing whether ?opioid receptor in the superficial layers of the rat spinal cord is located in the primary afferent C fiber or not. Materials and Methods: Based on the selective binding of Griffonia simplicifolia isolection I-B4(I-B4) to primary afferent C fiber, the I-B4 binding sites and opioid receptor (DOR) in the superficial layers of the rat spinal dorsal horn were labeled by double (immunohistochemical ) histochemical electron microscopy(ABC-immunogold method). Reuults: Under electron microscope, a lot of axons and axonal terminals were found immunoreactive to DOR. Moreover, some of I-B4 binding site-positive axons and axonal terminals were found DOR-immunoreactive in the double-labelled sections. Conclusion; DORs in the superficial layers of the spinal dorsal horn are partially localized in the primary afferent C fibers, which is involved in the processing of nociceptive afferent information thus to mediate the presynaptic inhibition of opioids, such as enkephalin on primary afferent C fiber.
