摘 要:为研究活性维生素D3对大鼠系膜增生性肾炎Caspase-3表达的影响,探讨1,25-二羟维生素D3对系膜增生性肾炎作用的可能机制。将清洁SD级大鼠随机分为对照组和肾炎模型组,后者随机分为肾炎组和活性维生素D3干预组,检测其肾小球及肾间质中Caspase-3的表达,进行比较分析。结果显示,1.Caspase-3在正常肾组织的肾小球中无表达,与对照组比,肾炎组中Caspase-3在肾小球系膜区表达增多,P<0.01;活性维生素D3组Caspase-3表达略高于肾炎组,差异无统计学意义,P>0.05;2.Caspase-3在正常肾小管上皮细胞中有少量表达,与对照组相比,肾炎组肾近端小管及远端小管随时间延续,表达明显增多,差异有统计学意义,P<0.01;给予活性维生素D3干预后3d,Caspase-3的表达高于肾炎组,但是到了7d与14d明显低于肾炎组,差异有统计学意义,P<0.05。由此可知,活性维生素D3可能参与了对Caspase-3的调节,诱导系膜细胞凋亡,促进肾炎的修复;减少大鼠肾小管上皮细胞凋亡,防止肾纤维化,保护肾脏,延缓肾脏疾病进展。To investigate the effect and mechanism of 1,25-dihydroxyvitamin D3 on Caspase-3 expression of rat mesangial proliferative glomererulonephritis.Methods:Clean SD rats were randomly divided into control group and the glomerulonephritis model groups,which was subdivided into the glomerulonephritis group and 1,25-(OH)2D3 group.Immune-histochemistry was used to detect the expression of Caspase-3 in glomerular and renal interstitium,to compare the differences between the groups.Results:1)There was no expression of Caspase-3 in renal glomeruli,in the glomerulonephritis group,Caspase-3 presented expression in Mesangium,P0.01;compared with the control group,the expression of Caspase-3 in 1,25-(OH)2D3 group was improved,but the difference was not statistically significant,P0.05;2)Caspase-3 in normal renal tubular epithelial cells had a small amount of expression.compared with the control group,Caspase-3 of the glomerulonephritis group in renal proximal tubules and distal tubules with time continuation of expression was significantly increased,and the difference was statistically significant(P0.01);Given to the intervention of 1,25-(OH)2D3,after three days,Caspase-3 expression became higher than the glomerulonephritis group,but after 7 days and 14 days treatment,the expression was significantly lower than nephritis group,with statistically significant difference(P0.05).1,25-(OH)2D3 may participate in the regulation of caspase-3,then enhance inflammation recoveredy;but it may reduce renal tubule epithelial cells apoptosis in rat MsPGN model,prevent renal fibrosis,and delay the progression of rat renal glomerular disease.
