摘 要:为研究大孔吸附树脂分离纯化洋葱总黄酮的工艺条件及参数。以洋葱总黄酮吸附率与解析率为指标,考察了2种不同型号的大孔树脂对洋葱总黄酮的吸附和解吸附能力,确定AB-8为最佳树脂,并对AB-8大孔树脂的纯化条件进行优化。结果显示,最佳工艺条件为pH=4、1~2mg/mL的浓度上样,上样流速为1.5BV/h,样品上样量为3mL/g树脂;用60%乙醇洗脱。经AB-8树脂处理后的总黄酮纯度达69.8%,收率为86.8%。由此可知,AB-8树脂对洋葱总黄酮的吸附量大,解吸附容易,可以应用于洋葱总黄酮的分离纯化。The process parameters of the separation and purification of onion total flavonoids with macroporous adsorption resins were studied.Two kinds of macroporous adsorption resins were studied on the static capacity of adsorption and desorption,and the better resin was AB-8.The optimum adsorption and desorption parameters of purification onion total flavonoids with resin AB-8 were as follows:pH=4,adsorption concentration of total flavonoids is 1~2 mg/mL,flow rate 1.5BV/h,quantity of the sample is 3mL/g,elution solvent 60% alcohol for desorption.The purification of onion total flavonoids with macroporous adsorption resins AB-8 was about 69.8%,and percent yield was 86.8%.The results show that resin AB-8 has good ability of purification of onion total flavonoids,and it could be used to separate and purify onion total flavonoids.
