松质骨成骨细胞的培养和鉴定

摘　　要：目的:探讨理想的人松质骨成骨细胞体外培养的方法。方法:取人髂骨松质骨,剪碎,用改良的酶消化法分离培养人成骨细胞;从细胞的形态学、增殖、碱性磷酸酶染色及矿化结节茜素红染色等方法鉴定培养的成骨细胞。结果:分离培养的人成骨细胞生长状态良好,纯度较高,具有典型的成骨细胞形态特征,碱性磷酸酶染色及矿化结节茜素红染色阳性。结论:改良的酶消化法分离培养松质骨可获得大量纯度较高的人成骨细胞。Objective： To investigate an ideal method of culturing human osteoblasts from cancellous bone in vitro.Methods： Human osteoblasts were isolated from cancellous bone of ilium by improved enzymatic digestion and identified by cell morphology,proliferation,alkaline phosphatase staining and mineralized nodules with alizarin red staining.Results： The growth of human osteoblasts was in good condition and high purity,with typical morphological features of osteoblast,alkaline phosphatase staining and mineralized nodules alizarin red staining were positive.Conclusion： High purity of human osteoblasts are derived from cancellous bone by improved enzyme digestion.
