单唾液酸神经节苷脂对体外循环后大鼠海马细胞超微结构的影响

摘  要:目的:探讨单唾液酸神经节苷脂(GM1)对大鼠体外循环(CPB)后海马细胞的超微结构的影响。方法:选用雄性SD大鼠,经右颈静脉插管引流,尾动脉插管灌注建立CPB,转流时间l h,建立CPB动物模型。随机分为CPB模型组、CPB模型+GM1组、输血组和假手术组,每组10只。采用透射电镜技术观测海马细胞的超微结构。结果:CPB后神经细胞出现粗面内质网和游离核糖体明显减少,线粒体肿胀,空泡化,嵴断裂,溶解,消失,细胞膜崩解;神经丝模糊、突触间隙模糊不清,出现较多神经细胞凋亡。GM1干预后神经细胞的超微结构轻度改善,突触间隙增宽,凋亡细胞减少。结论:CPB可导致大鼠海马神经元和突触的超微结构病变;GM1可通过减轻CPB后脑细胞超微结构的损害。Objective: To explore the protective effects of monosialoteterahexosyl ganglioside 1 (GM1) on brain damage after cardiopulmonary bypass (CPB). Methods: SPF-class adult male SD rats were randomly divided into CPB, CPB+ GM1, transfusion and sham-operated groups. After establishment of rat CPB model, GM1 of 20 mg/kg was added to the priming solution. Electron microscopy was used to observe the uhrastructure of the hippocampus, especially the structural parameters of synaptic interface changes. Resuits: After CPB, neural cell rough endoplasmic reticuium and free ribosomes were markedly decreased, mitochondriosome was swelled, vacuolated and destructed, cristae was ruptured, dissolved and disappeared, cellular membrane disintegrated; synaptic cleft was obviously vague, and neuronal apoptosis occur more frequently. Compared with the CPB group, the intervention of GM1 could slightly improve the above morphological changes, slightly narrowing synaptic cleft, and apoptotic cells decreased. Conclusion: CPB can lead to uhrastructure change of the neuron and synapse on hippocampus in rats. GM1 may attenuate cerebral uhrastructure injury associated with CPB.
