摘  要:目的观察促黑素对小鼠血清骨特异性碱性磷酸酶(ALP-B)和I型胶原交联C端肽(CTX-I)含量的影响。方法 9周龄雄性C57BL/6小鼠,随机分为对照组和实验组(每组8只),分别皮下注射生理盐水0.2 mL和促黑素100 nmol(加生理盐水0.2 mL),注射1,3,5 d后,摘眼球取血,测定血清ALP-B和CTX-I的含量。体外培养原代成骨细胞,用RT-PCR法测定黑皮质素受体(MCR)的表达。结果皮下注射促黑素3,5 d后,实验组与对照组相比,血清的ALP-B含量显著减少而CTX-I含量显著增加(均P<0.05)。小鼠原代成骨细胞表达MC1R~2R、MC,4R~5R。结论促黑素可能通过MCR作用于成骨细胞和破骨细胞对小鼠血清ALP-B和CTX-I产生影响。Objective To investigate the effect of a - melanocyte stimulating hormone (a -MSH ) on mouse serum bonespecific alkaline phosphatase (ALP- B ) and C -terminal cross- linking telopeptide of type 1 collagen ( CTX - I). Methods a - MSH were subcutaneously injected into 9 - week C57BL/6 male mice , and the content of serum ALP - B and CTX - I were measured 1, 3, 5 day after treatment. Melanocortin receptor(MCR) subtypes in the primary osteoblasts were measured by RT - PCR. Results The content of serum ALP - B were significantly decreased in 3 and 5 days after treatment with a - MSH (P 〈 0. 05). The content of serum CTX - I were significantly increased in 3 and 5 days after treatment (P 〈0. 05). MC,1R-2R,MC,4R -5R were expressed in primary cultured osteoblasts. Conclusion a - MSH might affect the content of ALP - B and CTX - I in mouse serum through MCRs on osteoblasts and osteoelasts.
