高效液相色谱法测定养殖水中的总氮

摘　　要：建立了适合养殖水中总氮的高效液相色谱测定方法。方法采用Hypersil ODS-C_(18)(5μm,250mm×416mm)色谱柱,流动相乙腈-水(20:80,V/V)体系,流速为0.20mL/min,紫外检测波长为204nm。校准曲线回归方程为A=5189C(mg/L)+7195,决定系数(R^2)为0.9996,方法检出限达0.02mg/L。方法应用于实际样品养殖水中总氮的测定,5次平行测定相对标准偏差(RSD)为1.09%—5.47%,加标回收率为91.0%—98.6%。A new high-performance liquid chromatography（HPLC） method was established to determine total nitrogen in aquaculture water.A Hypersil ODS-C_（18）（5μm,125mm X 416mm） column was selected as the chromatographic column with mobile phase of acetonitrile-H_2O（20：80,V/V） at a flow rate of 0.20mL/min,UV detection wavelength was 204nm.The regression equation was A=5189C（mg/L） ＋ 7195,the coefficient of determination was 0.9996.The detection limit was 0.02mg/L,the recoveries were 91.0%—98.6%.
