摘 要:目的探讨DNA酶Ⅰ(DNaseⅠ)基因多态性在新疆阿克苏地区维族人群中的分布及其与血脂、血糖的关系。方法选取新疆阿克苏地区维族健康体检者148例,提取外周血基因组DNA,通过高温连接酶检测法(PCR-LDR)检测DNaseⅠ基因A2317G(rs1053874)位点的基因型。酶法检测血清TC、TG及FPG水平;选择性遮蔽法测定LDL-C、HDL-C水平。结果 A2317G位点AA、AG、GG基因型在新疆阿克苏地区维族健康人群中分布频率分别为20.95%、54.05%和25.00%,A、G等位基因频率分别47.97%、52.03%,这与其在中国汉族人群中的分布频率相似(P>0.05),与蒙古人、日本人、韩国人、土耳其人、纳米比亚人、德国人明显不同(P<0.05)。3组基因型(AA/AG/GG)的血清TC、TG、LDL-C、FPG水平无统计学差异(P>0.05),而HDL-C水平差异有统计学意义(P<0.05)。结论 DNaseⅠ基因多态性具有种族异质性;在新疆阿克苏地区维族人群中其多态性与HDL-C水平有相关性。ObjecUve To investigate the frequency of deoxyribonuclease I gene polymorphism in Uygur Nationality of Aksu ar- ea in Xinjiang and it's association with blood glucose and lipid. Methods Blood samples of 148 healthy Uygur Nationality of Aksu area in Xinjiang were collected. Genomic DNA were extracted from all blood samples,PCR-LDR reaction was employed to detect the A2317G(rs1053874) mutation of deoxyribonuclease I gene,genotype and allele distribution were analyzed. Meantime, blood glu- cose and lipid were measured in fasting state by biochemical methods. Results Three genotypes of A2317G,including AA,AG and GG, were detected in Uygur Nationality of Aksu area in Xinjiang, the frequencies were 20.95 %, 54.05 % ,25.00 % respectively. The allele frequencies of A and G were 47.97 % and 52.03 % respectively, which were similar to the distribution frequency in the Chi- nese Han population(P〉0.05) and different from Mongolian,Japanese, Korean, Turks, Namibia and German(P〈0.05). Genotypes o( AZR17G were not associated. with serum total cholesteroL, triglyceride, low density lipoprotein-cholesterol and fasting glucose, but were closely related to high-density lipoprotein cholesterol(P〈0.05). Conclusion The frequency of deoxyribonuclease I gene pol- ymorphism could vary from different races,in Uygur Nationality of Aksu area in Xinjiang the genotypes were associated with high- density lipoprotein cholesterol.
