摘 要:目的:观察栀子豉汤对自发性高血压大鼠(SHR)的血压和血管紧张素Ⅱ(AngⅡ)1型受体(AT1R)mRNA表达的影响,初步探讨其防治糖尿病心血管病变的可能机制。方法:选取SHR 50只,按血压随机分为模型对照组,栀子豉汤4 g生药/kg、8 g生药/kg、16 g生药/kg组及卡托普利组(2 mg/kg),采用鼠尾动脉间接测压法测定大鼠血压,逆转录-聚合酶链式反应(RT-PCR)检测主动脉及心肌组织AT1R mRNA的表达。结果:栀子豉汤8 g生药/kg及16 g生药/kg组治疗4周后,SHR血压明显降低,主动脉及心肌AT1R mRNA表达下调。结论:栀子豉汤可降低血压,下调主动脉及心肌组织AT1R mRNA表达,可能对糖尿病心血管病变的防治具有一定价值。To observe the effects of Gardeniae and Sojae Praeparati Decoction (GSPD) on blood pressure (BP) and the expression of Angiotensin II type 1 receptor (AT1R) mRNA of spontaneously hypertensive rats (SHR) and to preliminarily explore its possible mechanism of preventing diabetic cardiovascular disease. Methods: 50 SHR were randomly divided into model group, GSPD 4 g crude drug/kg, 8 g crude drug/kg, 16 g crude drug/kg group and captopril group (2 mg/kg) accord!ng to BP. The indirect tail-artery blood pressure measure- ment was used to measure BP, the expression of AT1R mRNA in aortic and myocardium tissue was detected by reverse transcription polymer- ase chain reaction (RT-PCR) method. Results: After GSPD 8 g crude drug/kg and 16 g crude drug/kg group treated for 4 weeks, the BP of SHR was decreased significantly, the expression of AT1R mRNA in aortic and myocardium was down-regulated. Conclusions: GSPD can de- crease BP, down-regulate the expression of AT1 R n1RNA in aortic and myocardium tissue, thus .GSPD may have potentially clinical values in the prevention and treatment of diabetic cardiovascular disease.
