摘 要:目的:研究黄连解毒汤中黄酮成分对人类红白血病耐药细胞(K562/ADM细胞)的增殖抑制作用及相关因子表达的影响,并对其作用机制进行初步的探讨。方法:采用MTT法检测黄连解毒汤中黄酮成分对细胞增殖抑制的影响,半定量RT-PCR法检测mdr-1,TopoⅡ及Caspase-3基因的表达。结果:10μg/ml黄芩苷、20μg/ml京尼平苷对增值抑制作用明显,PCR结果显示50μg/ml黄芩苷、100μg/ml京尼平苷均可以通过降低mdr-1、TopoⅡ的表达,升高Caspase-3基因的表达,发挥逆转白血病细胞MDR的作用。结论:黄连解毒汤中黄芩苷、京尼平苷为其干预与逆转肿瘤多药耐药的有效成分,其作用机制可能与改变mdr-1、TopoⅡ及Caspase-3基因的表达有关。To investigate the reversal effect of the flavonoids ingredients extract from Huanglianjiedu Decoction on proliferation inhibi- tion of MDR of myelo genous leukemia K562/ADM cells and on expression of K562/ADM cell~ multidrug resistance related gene, discuss the possible correlative mechanism. Methods: The inhibitory rates of these cells were measured by MTr assay and define the effective chemi- cal constituents. The mdr-1 ,Topo II and Caspase-3 were measured with RT-PCR. Results: Baicalin, geniposide could inhibit the growth of K562/ADM cell,the relation of dosage-effct. Baicaln,geniposide can decrease expression of mdr-1 and Topo II ,at the same time it can in- crease expression of and Caspase-3. Conclusion: Defined baicaln, geniposide is effective chemical eonsituents at work, its mechanism may be related to down- regulating of the mdr-1 ,Topo II and Caspase-3 expresion.
