摘  要:目的:研究雷公藤红素对HER2/neu过表达人类乳腺癌MDA-MB-453细胞周期的影响。方法:流式PI单染检测细胞周期的分布;Western Blot检测细胞周期相关蛋白的改变。结果:用3.00、6.00、12.00μM雷公藤红素处理MDA-MB-453细胞24 h后,细胞周期阻滞在G0/G1期,并检测到该周期相关蛋白Cyclin D1、P27、p-Rb均发生了变化,且呈浓度依赖性改变。结论:雷公藤红素诱导MDA-MB-453细胞周期停止于G0/G1期,且与P27上调,Cyclin D1蛋白水平下调及Rb的高磷酸化状态下降有关。To study the Effects of eelastrol on the cell cycle of MDA-blB-453 cancer cells. Methods: The cell cycle distribution was tested by flow cytometry; The expression of cell cycle related proteins were determined by western blot. Results: After MDA-MB-453 cells were treated with 3.00 - 12.001μM of celastrol for 24 h, cells were arrested in G0/G1 phase, and the expression of G0/G1 related proteins Cyelin D1, P27 and p-Rb were changed. Conclusion: Celastrol induced G0/G1 arrest in Her2/neu-overexpressing MDA-MB-453 cancer cells. This G0/G1 arrest was associated with decreasing the protein expression of Cyclin D1 ,p-Rb and increasing of P27 protein.
