摘 要:目的:观察多酚类化合物芒果苷对四氧嘧啶性糖尿病小鼠糖脂代谢的调节作用,并探讨其作用机制。方法:采用雄性ICR小鼠尾静脉一次性注射四氧嘧啶(60.0 mg/kg)诱导糖尿病小鼠动物模型。芒果苷(12.5、25、50mg/kg)腹腔注射给药4周,测定各组糖尿病小鼠血糖、血脂、糖原和血清胰岛素的水平;通过对小鼠胰腺的H.E染色和胰岛B细胞的醛品红染色,观察芒果苷对胰岛和B细胞的影响。结果:芒果苷腹腔注射给药4周后,25、50mg/kg芒果苷组能显著降低糖尿病小鼠血糖,并增加其肌、肝糖原含量;50mg/kg芒果苷组糖尿病小鼠血清胰岛素水平、胰岛和B细胞的数量显著高于糖尿病模型组。12.5、25、50mg/kg芒果苷能显著降低糖尿病小鼠血清甘油三酯和总胆固醇含量。结论:芒果苷通过促进糖尿病小鼠肌、肝糖原的合成和胰岛素的分泌,降低血清甘油三酯和总胆固醇含量来调节糖尿病小鼠的糖脂代谢。Mangiferin (MF) is a polyphenol compound, the present study was carried out to investigate the effect and mechanism of mangiferin on glucose and lipids metabolism in alloxan-induced diabetic mice. Methods: Adult male ICR mice were injected alloxan 60 rag/ kg body weight though tail vein to induce diabetic animals. The effects of mangiferin on glucose and lipids metabolism were investigated in al- loxan-induced diabetic mice. Mangiferin( 12.5,25.0,50. Omg/kg) was administered intraperitoneally (ip) for four weeks. At the end of ex- periment, blood glucose, blood lipids and serum insulin were determined,the glycogen content of hepatic and skeletal muscle were also meas- ured. The pancreatic islet and B-cell were identified by stain with hematoxylin eosin (HE) and aldehyde-fuchsin. Results: After 4 weeks ad- ministration of mangiferin,mangfferin (25.0,50.0 mg/kg) significantly lowered blood glucose ,and greatly increased muscle and hepatic gly- cogen content, compared with diabetic mice( P 〈 0.05 ) . The intraperitoneal administration of mangiferin (50.0 mg/kg) showed significant elevation of insulin in serum, and increased the number of pancreatic islet and B cell. , compared with diabetic mice ( P 〈 0.05 ). Mangiferin (12.5,25.0,50.0mg/kg) significan-tly declined the level of triglyceride and total cholesterol in a dose-dependent manner, compared with di- abetic mice( P 〈 0.05) . Conclusion: mangiferin have some modulating effects on glucose and lipids metabolism in alloxan-induced diabetic mice,which mechanism was related to increase glycogen and serum insulin, decrease triglyceride and total cholesterol in the serum.
