摘 要:目的:观察5种经研究有抗肝纤维化作用的中药成分:灯盏花素、青蒿琥酯、红景天苷、黄芪总甙、粉防已碱对肝星状细胞(HSCs)收缩的影响,筛选出能抑制HSCs收缩的中药成分。方法:分离培养大鼠HSCs,通过MTT及细胞形态观察研究100~0.1μmol/L灯盏花素、青蒿琥酯、红景天苷、黄芪总甙、粉防已碱对HSCs增殖及毒性作用,以10nmol/L内皮素(ET-1)为刺激因子,用凝胶收缩实验观察各中药成分对ET-1刺激下HSCs收缩的影响,筛出能抑制HSCs收缩的成分,凝胶面积用"%原始面积"表示。结果:100~0.1μmol/L灯盏花素、青蒿琥酯、黄芪总甙、粉防已碱具有浓度依赖性地抑制HSC增殖的作用,其中100μmol/L黄芪总甙,100μmol/L及10μmol/L粉防己碱对HSCs有毒性作用。100μmol/L及10μmol/L灯盏花素凝胶面积分别为74.33±5.5%和67.39±3.47%,与ET-1组面积(49.96±4.46%)相比有显著差异(P﹤0.01);100μmol/L及10μmol/L青蒿琥酯凝胶面积分别为79.62±1.95%和58.86±2.99%,与ET-1组面积(39.85±3.12%)相比有显著差异(P﹤0.01)。100~0.1μmol/L红景天苷、10~0.1μmol/L黄芪总甙和1及0.1μmol/L粉防己碱凝胶面积与ET-1组相比无明显差异。结论:5种中药成分中,100及10μmol/L灯盏花素及青蒿琥酯均能抑制ET-1诱导的大鼠HSCs收缩。To screen ingredients of traditional Chinese medicine with effect on inhibiting contraction of hepatic steUate cells (HSCs) induced by endothelin-1 among breviscapine, artesunate, salidroside, astragalosides and tetrandrine. Methods: HSCs were isolated from Sprague-Dawley male rats. MTT method was used to study the effect of above 5 ingredients on proliferation of HSCs with the concentration of 100,10,1 and 0.1 μmol/L. In addition, collagen gel contraction assay was used to study the effect of above 5 ingredients on inhibiting con- traction of HSCs induced by 10nmol/L ET-1. Results: The proliferation of HSCs was inhibited by 100 ± 0.1 panol/L breviscapine, artesu- hate, astragalosides and tetrandrine in a concentrati0n-dependent manner. Toxicity effects were observed after 100μmol astragalosides, and 100lμmol/L or 10μmol/L tetrandrine were added. The contraction of HSCs induced by 10nmol/L ET-1 was suppressed by pre-incubated with 100μnol/L or 101μmol/L breviscapine. The area of collagen gel was 74.33 ± 5.5% or 67.39 ± 3.47% respectively ( vs. 49.96 ±4.46% in ET-1 group, both P 〈 0.01 ). Besides, the contraction of HSCs was also suppressed by pre-incubated with 100tunol/L or 10pznol/L artesu- nate. The area of collagen gel was 79.62% ± I. 95% or 58.86% ± 2.99% respectively ( vs. 39.85 ± 3.12% in ET-1 group, both P 〈 0. 01 ). However, the contraction of HSCs was not significantly suppressed by per-incubated with 100 ± 0. 1 μmol/L salidroside, 10 ± 0. 1 μnol/ L astragalosides, or 1 and 0. 1 μmol/L tetrandfine, respectively. Conclusions: 100 and 10μnol/L breviscapine and artesunate could inhibit contraction of HSCs induced by ET-1.
