摘 要:目的:研究甘草总黄酮(FEPs)的抗抑郁作用。方法:雄性SD大鼠正常组、模型组、甘草黄酮组(30、100、300mg/kg)、阳性药物氟西汀组各10只,采用成年大鼠慢性不可预测温和应激模型(CUMS),连续给药28天后,采用旷场实验(OFT)、强迫游泳实验(FST)、悬尾实验(TST),并实施血清皮质酮检测,判断甘草总黄酮的抗抑郁作用。雄性昆明小鼠正常组、甘草黄酮组(5、30、180mg/kg)、阳性药物组氟西汀各16只,连续21天给药,并在实验第1、7、21天进行自主活动实验监测,观察FEPs的中枢作用特点。结果:与模型组相比较,甘草总黄酮(30、100、300mg/kg)干预后,大鼠在旷场实验中表现直立次数增多、穿格数增多、粪便粒数明显减少;强迫游泳不动时间明显减少;悬尾实验不动时间明显减少;甘草黄酮300mg/kg剂量干预后血清皮质酮明显低于模型组;甘草总黄酮对供试小鼠无中枢兴奋作用。结论:甘草总黄酮具有抗大鼠慢性不可预测温和应激模型引起的行为抑郁作用,且无中枢兴奋的不良反应。To study the antidepressant effect of the flavonoids extracted parts (FEPs) from Glycyrrhiza uralensis Fisch. Method: 60 male Sprague-Dawley (SD) rats were randomly assigned into 6 groups as control, model ( stress + vehicle, po), fluoxetine group ( stress + fluoxetine 3.5 mg/kg, po) and 3 FEPs groups (stress + FEPs 30, 100, 300mg/kg, po). All rats were exposed to chronic unpredictable mild stress (CUMS) for 28 consecutive days. Forced Swimming Test, Open Field Test, Tail Suspension Test were applied to assess the anti- depressant efficacy, and the serum corticosterone level was measured by radio immunoassay method. We still performance the locomotor activ- ity test to evaluated the central never system property of the flavonoids by randomly dividing 96 male Kunming mouse into 6 groups as normal control group, diazepam group (5mg/kg, po), fluoxetine group (fluoxetine 20mg/kg, po) and 3 FEPs groups (5,30,180mg/kg. po). Re- sult: The flavonoids extracted parts from Glycyrrhiza uraleusis can significantly short the motionless time in Forced Swimming Test ( P 〈 0. 05, P 〈 0.01 ) and in Tail Suspension Test ( P 〈 0.05, P 〈 0.01 ). In the Open Field Test, the flavonoids can improved the stress behavior performance by increasing rearing times ( P 〈 0.05, P 〈 0.01 ) and the horizontal crossing numbers ( P 〈 0.01 ), and by decreasing the num- ber of number of fecal boli produced ( Defecation ) ( P 〈0.01 ) . Still, the 300mg/kg dosage of flavunoids can significantly lower the serum corticosterone level in CUMS rats (P 〈 0.01 ) and the flavonoids have no effect on the locomotor activity test in mice. Conclusion: The fla- vunoids extracted parts from Glycyrrhiza uralensis in Ningxia showed the antidepressant effect on CUMS rats and without the central excitatory effect in mice.
