摘 要:目的:观察痰瘀同治方对高脂血症MI/RI大鼠血脂及代谢产物谱的影响。方法:30只雄性SD大鼠随机分为假手术组、模型组及痰瘀同治组3组。除假手术组外,其余大鼠给予高脂饲料喂养,采用可逆性冠脉左前降支结扎缺血30min再灌注2h复制MI/RI模型,运用核磁共振的代谢组学技术和生化分析技术,观察痰瘀同治方干预下,高脂血症MI/RI大鼠血脂及代谢产物谱的变化情况。结果:与假手术组比较,模型组大鼠血清CHO、TG、LDL-L显著升高且HDL-L含量降低(P<0.01或P<0.05);与模型组比较,痰瘀同治组能明显降低CHO、TG含量(P<0.05)。主成分分析法能将假手术组、模型组及痰瘀同治组明显区分开,不同组别之间代谢产物存在差异,给予痰瘀同治方后可以干预代谢物而致代谢终产物的改变。结论:高脂血症MI/RI大鼠血浆代谢组学出现变化,痰瘀同治方能够部分调控高脂血症MI/RI大鼠血浆代谢中发生异常的代谢产物,对其质代谢紊乱有一定促进恢复的作用。Objective:To investigate the effects of Tanyu Tongzhi(TYTZ) recipe on blood lipid and metabolic profiles of hyperlipidemia and myocardial ischemia-reperfusion injury in rats. Method:30 SD male rats were divided into 3 groups randomly, sham-operated group, model group and the TYTZ group. All rats were fed with high-fat diet expect the shamoperated group. The model of MI/RI myocardium was reproduced by ligation of left descending artery for 30 min followed by releasing the ligation for 2 hours in rats. The changes in blood lipid and metabolic profiles of the model rats were then observed using biochemical methods and NMR-based metabonomic techniques. Results. Compared with sham-operated group, the levels of CHO, TG, LDL-L, were significantly higher ,however, HDL-L were lower in model group(P 〈 0. 01 or P 〈0. 05). CHO and TG were obviously lower in TYTZ group than the model group(P 〈0. 05). The metabolic profiles of the model rats were different from the sham-operated group and TYTZ group, and the metabolites of different groups were clearly distinguished. TYTZ recipe could intervene the metabolites or the metabolic path to cause changes in final metabolites. Conclusion: The metabonomics chianges in rats of stagnation of hyperlipidemia and myocardial ischemiareperfusion injury, and TYTZ recipe can regulate the disturbance and make it incline to the normal state.
