摘  要:目的:探讨蛋白质组学在大肠癌气血方虚证中的研究情况,筛选大肠癌气血亏虚证、瘀血证及平和体质大肠癌患者血清差异表达的蛋白质。方法:收集大肠癌姑息期气血亏虚证、瘀血证和大肠癌平和体质患者外周血清各6例,通过双向凝胶电泳以及质谱鉴定和生物信息学分析,寻找在大肠癌姑息期气血亏虚证患者血清中特异表达的蛋白质。结果:选择表达差异明显的3个蛋白质点进行质谱鉴定和生物信息学分析,其中的白细胞介素-8(IL-8)、纤维结合蛋白在大肠癌姑息期气血亏虚证患者血清中均为高表达;血清载脂蛋白A-I前体在大肠癌姑息期气血亏虚血清中为低表达。结论:三种蛋白差异表达蛋白质的组合性变化可能是大肠癌气血亏虚证的物质基础,具有潜在的诊断、预后标志物或治疗靶点的意义。Objective: To screen serum differentially expressed proteins for colorectal cancer patient of qi and blood deficiency syndrome,blood stasis syndrome and mild constitution.Methods: Peripheral serums of colorectal cancer patient of qi and blood deficiency syndrome,blood stasis syndrome and mild constitution in palliative period were collected each with six cases.Serum differentially expressed proteins for colorectal cancer patients of qi and blood deficiency syndrome were looked for by two-dimensional gel electrophoresis,mass spectrometry and bioinformatics analysis.Results: Three protein points with apparent expression differences were collected to identify by mass spectrometry,and analysis of bioinformatics,and interleukin-8,fibronectin of whose were all highly expressed for serums of colorectal cancer patients of qi and blood deficiency syndrome;while apolipoprotein A-I precursor were decreased in the same conditions.Conclusion: Combinational changes of three kinds of proteins of differential expression may be the material bases for colorectal cancer of qi and blood deficiency syndrome,which has the significances of potential diagnosis,prognosis markers or therapeutic targets.
