摘 要:目的:建立顽痹清丸的质量标准。方法:用薄层色谱法对顽痹清丸中络石藤、乳香、牡丹皮、益母草进行定性鉴别,用高效液相色谱法测定顽痹清丸中黄芩苷的含量,采用色谱柱Agilent C18柱(4.6 mm×250 mm,5μm);流动相:甲醇-0.1%磷酸溶液(47∶53);流速:1.0 mL·min-1;柱温:30℃;检测波长为280 nm。理论塔板数以黄芩苷计不低于5 000。结果:薄层色谱鉴别法能定性鉴别络石藤、乳香、牡丹皮、益母草,斑点清晰,分离度好,专属性强,且阴性无干扰。含量测定中黄芩苷在0.277 6~2.776 0μg范围内呈良好的线性关系,r=0.999 4;平均加样回收率为99.10%,RSD为1.29%。结论:该方法简便可行、结果准确、重现性好,可作为顽痹清丸的质量标准。Objective: To establish the quality standards of Wanbiqing Pill.Methods: Thin layer chromatography(TLC) was applied for the qualitative identification of Starjasmine Stem,Frankincense,Cortex moutan,and Herba Leonuri in Wanbiqing Pill and HPLC method was used to determine the content of baicalin in Wanbiqing Pill.Agilent C18 Column(4.6 mm × 250 mm,5 μm) was used as chromatographic column,and methanol-0.1% phosphoric acid(47∶ 53) as the mobile phase.The flow rate was 1.0 mL·min-1 and the column temperature was 30 ℃.The detection wavelength was 280 nm.The number of theoretic plate of baicalin was not less than 5 000.Results: Starjasmine Stem,Frankincense,Cortex moutan,and Herba Leonuri could be qualitatively identificated by TLC,which had clear spots,good separating degree,strong specificity,and negativity without interference.The content determination of baicalin had good linear relation in the range of 0.277 6-2.776 0 μg,r = 0.999 4;the average recovery rate was 99.10%,and RSD was 1.29%.Conclusion: This method is simple and feasible,accurate,reproducible,which can be regarded as the quality control of Wanbiqing Pill.
