摘 要:目的研究米诺环素在体内外干扰骨髓间充质干细胞后对其生长的影响。方法大鼠脊髓损伤后,经口给予米诺环素,并将带有GFP基因的SD大鼠骨髓间充质干细胞局部移植到损伤脊髓,在体外培养大鼠骨髓间充质干细胞并给予不同剂量的米诺环素进行干预。利用荧光显微镜计数细胞,采用MTT法描绘细胞生长曲线来观察干细胞的生长状况。结果米诺环素联合骨髓间充质干细胞治疗实验性脊髓损伤,在损伤中心可以观察到更多的移植细胞,体外培养的骨髓间充质干细胞在合适剂量米诺环素的干预下呈现出更加旺盛的生长趋势。结论合适剂量的米诺环素可以促进体内移植的骨髓间充质干细胞在损伤脊髓内更多地迁移到损伤中心,而对于体外培养的骨髓间充质干细胞米诺环素会促进其增殖。Objective To study of the influence of minocycline microenvironment on bone marrow mesenchymal stem cells (BMSC) both in vitro and in vivo. Methods The spinal cord injured rats were given minocycline by oral administration, BMSC from the GFP-SD rats were locally transplanted to the injury spinal cord;rat bone marrow mesenchymal stem ceils were also cultured in vitro and given different dose of minocycline, the status of cell growth were observed using the fluorescence microscope and MTT assay. Results More transplanted ceils were observed in the injury center of the minocycline and BMSC combined treatment group, a vigorous intervention was also found with appropriate dose of minocycline in the cultured BMSC growth trend. Conclusion The appropriate dose of minocycline plays an extremely important role in BMSC value-added and migration both in vivo and in vitro.
