摘 要:目的:研究疏血通注射液(SXT)对大鼠脑缺血再灌注后缺血侧脑组织细胞间黏附分子-1(ICAM-1)、血管细胞黏附分子-1(VCAM-1)蛋白的表达及分布的影响。方法:建立大鼠大脑中动脉栓塞(MCAO)模型,在缺血2h、再灌注的同时,通过股静脉给予疏血通注射液(0.7mL/Kg)。分为假手术组(Sham组),模型组(Model组),疏血通注射液处理组(Model+SXT组)。给药处理24h后,取缺血侧脑组织,检测缺血侧脑组织中ICAM-1蛋白和VCAM-1蛋白的表达水平。用4%多聚甲醛固定脑组织,石蜡切片免疫组织化学法观察ICAM-1、VCAM-1的分布。结果:蛋白质印迹法和免疫组化显示,模型组缺血侧脑组织中黏附分子ICAM-1、VCAM-1蛋白表达显著增加,疏血通注射液抑制了两种黏附分子表达的增加。结论:疏血通注射液可以通过抑制大鼠脑缺血再灌注后脑组织ICAM-1和VCAM-1的表达,从而减轻脑缺血再灌注损伤。Objective: To investigate the influence of Shuxuetong Injection (SI) on the expression of vascular adhesion molecules in cerebral ischemia and reperfusion rats. Methods: Rats were randomly allocated to Sham operation group, model group and SI group. The rat model of cerebral ischemia and reperfusion was indeed by middle cerebral artery occlusion (MCAO), and SI (0.7mL/kg) was given to the model rats through femoral vein after 2-hour ischemia and reperfusion. Lateral ventricle tissue was taken for the detection of protein expression of adhesion molecules intercellular adhesion molecule 1 (I- CAM-l) and vascular cell adhesion molecule 1 (VCAM-1) with western blot 24h treatment after treatment. Immunohisto- chemistry was used to detect the distribution of ICAM-1 and VCAM-1 in the paraffin sections of rat brain tissue. Results: The results of Western blot and immunohistochemistry showed that ischemia and reperfusion induced the increase of I- CAM-1 and VCAM-1 protein expression in the lateral ventricle tissue, and SI intervention depressed their increase. Conclu- sion: SI shows an effect on inhibiting ischmia/reperfusion-induced up-regulation of adhesion molecules in rat brain tissues, which might contribute to its protective effect on cerebral ischemia and reperfusion.
