摘  要:目的观察艾灸足三里穴对亚急性衰老小鼠脑细胞染色体端粒长度的影响,探究艾灸抗衰老在基因水平的作用机制。方法将36只健康雄性ICR小鼠按随机数字表法分为空白组、衰老模型组(对照组)、衰老模型加艾灸治疗组(治疗组)3组。通过背部皮下注射D-半乳糖30天制造亚急性衰老模型;艾灸治疗组隔日艾灸足三里穴,双侧轮换灸,共15次。30天后对各组小鼠进行跳台实验以比较其智能,测量各组小鼠脾重指数、脑细胞染色体端粒长度及脑重指数。结果治疗组小鼠与其他2组比较,跳台实验成绩优于其他2组(P>0.05);脾重指数较高,端粒长度较长,差异亦均有统计学意义(P<0.05或P<0.01);治疗组小鼠脑重指数虽优于对照组,但差异无统计学意义(P>0.05)。结论艾灸足三里穴能明显延缓脑细胞染色体端粒的缩短,这可能是艾灸抗衰老基因水平作用机制之一。Objective To observe the influence of moxibustion at Zusanli (ST 36) for telomere length of chromosome in brain cell of subacute aging mice and explore action mechanism of moxibustion for anti-aging in genetic level. Methods Thirty-six healthy male ICR mice were divided into three groups according to random digital table: a blank group, an aging model group (control group), an aging model and moxibusition treatment group (treatment group). Subacute aging models were established by subcutaneous injection of D-galactose on the back for 30 days. In the treatment group, moxibustion was applied at Zusanli (ST 36) every other day, two sides in turn, and totally 15 times were required. After 30 days, intelhgenee level was measured with step-down test in each group. Besides the spleen index, telomere length of chromosome in brain cell and brain weight index in each group were measured. Results Compared with the other two groups, the treatment group had a better result in step-down test (P 〉0.05), and the spleen index was higher and the telomere length was longer, which both had significant differences in statistics (P 〈0.05 or P 〈0.01). The brain weight index in the treatment group was superior to that in the control group, which was not statistically significant(P 〉0.05). Conclusion Moxibnstion at Zusanli (ST 36) could apparently delay shortening of the chromosome telomere in brain cell, which might be one of the action mechanisms on moxibustion for anti-aging in genetic level.
