金娃麦×葡萄牙野燕麦远缘杂交后代的GISH研究

摘　　要：以金娃麦×葡萄牙野燕麦杂交后代金燕为试验材料,以地高辛标记的葡萄牙野燕麦基因组DNA为探针、中国春基因组DNA为封阻,对金燕-158的根尖染色体进行基因组原位杂交(GISH)分析,试验结果表明在金燕后代品系中均检测到燕麦染色质的存在,且不同品系间带有燕麦染色体片段数目不同,进一步证明金燕是金娃麦×葡萄牙野燕麦远缘杂交的后代。We used ＇Jinyan-158＇ ,the distant hybridization generations from Avena fatua and Jinwa Wheat as experiment materials. GISH with genomic DNA of A. fatua labeled by DIG as probe and genomic DNA of Jinwa Wheat as blocker was used to identify alien chromosome in root tip chromosomes of ＇ Jinyan＇ to analyze the main factors influencing GISH results to establish optimal GISH system. The results showed that the chromosomes of A. fatua could be detected from Jinwa Wheat and different lines showed different fragment numbers of A. fatua, and verified ＇ Jinyan＇ were the distant hybridization generations from A. fatua and Jinwa Wheat.
