摘  要:在pH 4.0 Britton-Robinson缓冲溶液中,室温条件下,刚果红和血红蛋白反应生成缔合微粒,使体系的共振散射信号明显增强,并在529 nm处有一最大的共振散射峰。在选定的条件下,血红蛋白质量浓度在0.060~1.680μg/mL范围内与ΔI呈良好的线性关系,据此建立了一种测定血红蛋白含量的新方法。该方法的线性回归方程为ΔI=39.24ρ-1.776,相关系数r为0.9998,检出限为7.0×10-3μg/mL,将方法应用于尿样中血红蛋白含量的检测,回收率为94.2%~95.2%。In pH 4. 0 Britton-Robinson buffer solution,congo red reacted with hemoglobin to form ionassociation complexes at room temperature, which greatly enhanced the resonance light scattering intensity of the system at the wave length of 529 nm. Under the selected conditions, hemoglobin concentration was linear with AI in the range of 0. 060 to 1. 680 ~g/mL. Based on this, a new method for the determination of trace amounts of hemoglobin was developed. The regression equation was AI = 39.24p - 1. 776, with a correlation coefficient of 0. 9998. The detection limit was 7.0 × 10^-3 μg/mL. The proposed method was applied to the analysis of hemoglobin in human urine samples, and the recoveries were in the range of 94.2% - 95.2%.
