摘  要:目的:探讨丹酚酸B(SaIB)对二磷酸腺苷(ADP)和凝血酶(Th)诱导的血小板聚集率以及释放的可溶性P选择素(sP-sel)和可溶性CD40L(sCD40L)的影响。方法:采集健康志愿者静脉血,制备富血小板血浆(PRP),以终浓度10μmol/L ADP和0.4 U/mL Th为诱导剂,测定血小板聚集率,分别以300μg/mL和600μg/mLSalB预孵育血小板,测定聚集抑制率。制备血小板悬液,分为静息组、激活组(以终浓度50μmol/L ADP和0.4U/mL Th为诱导剂激活)、SalB组(以终浓度300μg/mL和600μg/mL SalB预孵育,再以诱导剂激活)。采用ELISA方法检测各组上清液中sP-sel和sCD40L含量,比较sP-sel和sCD40L的相关性。结果:SalB可显著减低ADP和Th诱导的血小板聚集率,600μg/mL组聚集抑制率高于300μg/mL组(P<0.01)。与静息组比较,激活组sP-sel和sCD40L释放显著增加(P<0.01)。与激活组比较,SalB可使Th诱导的sP-sel和sCD40L释放及ADP诱导的sP-sel释放显著降低(P<0.01);但对ADP诱导的sCD40L释放没有影响,与激活组比较差异无统计学意义(P>0.05)。ADP和Th诱导时,sP-sel与sCD40L水平呈正相关(P<0.05,P<0.01),相关系数r_P>0.5。结论:SalB可降低体外血小板聚集率及活化产物sP-sel和sCD40L的释放,具有阻抑血小板活化、聚集与释放的作用。Objective:To explore the effect of Salvianolic acid B(SalB) on platelet aggregation,soluble P-selectin (sP-sel)and soluble CD40L(sCD40L) release by activated platelets which were respectively induced by adenosine diphosphate(ADP)and thrombin(Th).Methods:Platelet Rich Plasma(PRP) collected from healthy volunteers,was induced respectively by ADP(10μmol/L)and Th(0.4 U/mL)for platelet aggregation rate.Inhibition rate was assayed under the pre-incubation of SalB(300μg/mL,600μg/mL).PLT suspension liquid was divided into four groups:resting group,activating group[induced by ADP(50μmol/L) and Th(0.4 U/mL)]and SalB group(terminal concentration of 300μg/mL and 600μg/mL,pre-incubate and activated by inducers).sP-sel and sCD40L contents in each supernatant were assayed by ELISA to compare the correlation of sP-sel and sCD40L.Results:SalB can significantly reduce the rate of platelet aggregation by ADP and Th induction.SalB group(600μg/mL) inhibition rate was higher than that of 300μg/mL group(P0.01).Compared with resting group,release of sP-sel and sCD40L in activating group significantly increased(P0.01).Compared with activating group,SalB group(sP-sel and sCD40L induced by Th) and SalB group (sP-sel induced by ADP) were significantly reduced(P0.01);but the difference had no statistical significance between SalB group(sCD40L induced by ADP) and activating group(P0.05).The contents of sP-sel and sCD40L induced by ADP were positively correlated with those by Th(P0.05,P0.01),r_P0.5.Conclusions:SalB can reduce platelet aggregation rate in vitro and sP-sel and sCD40L which are released by activated platelets.SalB can inhibit platelet activation,aggregation and release.
