摘 要:目的:观察益气活血法中药对兔退变髓核中Fas/FasL的表达及基质代谢的影响。方法:建立兔腰椎间盘退变的动物模型,随机分为益气活血法中药治疗组和生理盐水治疗的对照组,治疗后用病理切片观察髓核的组织学变化;测量髓核中蛋白多糖含量;应用逆转录PCR法检测治疗后髓核中Fas/FasL基因mRNA表达。结果:①两组动物治疗后病理切片显示髓核退变均明显,但治疗组软骨终板退变较轻,血管芽较多;②治疗组1、2月和对照组1、2月蛋白多糖含量均减少,两组间对比无显著差异(P>0.05);各治疗组内1、2月蛋白多糖含量的组内对比(P>0.05),具有显著差异。③治疗组和对照组的随时间延长Fas/FasL基因mRNA表达均增强,治疗组和对照组对比无显著差异(P>0.05);各组组内对比Fas/FasL基因mRNA表达增强,具有显著差异(P<0.05)。结论:本实验提示益气活血法中药在一定程度上能延缓椎间盘软骨终板退变的发展,但不能根本抑制或逆转髓核形态学退变的发生;不能抑制髓核内Fas/FasL基因的表达;不能抑制基质中蛋白多糖含量的减少。Objective:To observe the influence of Yiqi Huoxue therapy on Faa/Fasl expression and matrix metabolism in nucleus pulpous degeneration of rabbit.Method:Established animal model of rabbits intervertebral disc degeneration, and they were divided into two groups:Yiqi Huoxue therapy group and the control group with normal saline.After treatments, the nucleus pulpous tissue was made into pathological section to observe the histological changes.The content of proteoglycan was tested and mRNA expression of Fas/FasL gene in nucleus pulpous was tested with reverse transcription PCR(polymerase chain reaction) after treatments.Results:①After treatments,the pathological section showed obvious degeneration of nucleus pulpous,but the cartilage endplate degenerated mildly in the treatment group.②The contents of proteoglycan in the two groups were all decreased in the first and second month.There was no difference between the two groups(P0.05 ).There were significant differences contrast in the group(P0.05 ).③The mRNA expression of Fas/ Fasl gene enhanced with time extension in the two groups,and there was no difference between the two groups(P0.05);there were significant differences contrast in the group(P0.05).Conclusion:The experiment shows that Yiqi Huoxue therapy with traditional Chinese medicine can delay the progress of intervertebral disc cartilage endplate degeneration, but cannot restrain or reverse the occurrence of nucleus pulpous morphological degeneration,cannot restrain the expression of Fas/FasL gene in nucleus pulpous and the decrease of proteoglycan content in matrix.
