摘 要:目的:对山核桃叶总黄酮的制备工艺进行了系统考察,为后继药效学研究奠定基础。方法:通过正交实验对黄酮提取工艺进行优化,以D101大孔树脂为吸附材料,对其静态吸附容量及黄酮洗脱富集条件进行考察,确定山核桃叶总黄酮制备工艺。结果:山核桃叶总黄酮的最佳提取工艺为:60%乙醇提取2次;提取时间分别为80 min、60 min;溶媒倍数分别为10倍、8倍。D101型树脂对山核桃叶总黄酮的静态吸附容量为(36.78±2.5)mg/g,洗脱次序为20%乙醇洗脱后收集40%乙醇洗脱部分即为山核桃叶总黄酮,其总黄酮得率为(74.93±2.10)%,纯度达(74.07±2.61)%。结论:所确立的山核桃叶总黄酮提取、纯化工艺合理、稳定,黄酮得率及纯度较高,为深入研究山核桃叶总黄酮奠定了良好基础。Objective:To prepare the total flavonoids of Carya cathayensis Sarg leaves(CCL) and settle foundation for its further pharmacology research.Methods:Orthogonal experimental design was applied to optimize the total flavonoids extract technology;static adsorptive capacity and flavonoids enriching condition were investigated on D101 macroporous resin. Results:The final optimized extraction technology was:CCL powder extracted with 10 and 8 times volume of 60%ethanol twice,with reflux time 80 min and 60 min respectively.Total flavonoids of CCL was prepared on D101 resin with static adsorptive capacity(36.78±2.5)mg/g;after eluting with 20%ethanol and collecting the 40%ethanol elute part,the total flavonoids yield was(74.93±2.10)%and with purity of(74.07±2.61)%detected by AlCl_3 colorimetric method. Conclusion:Data above indicated that the method established for preparation of the total flavonoids of CCL was rational and stable,which can pave the way for its further pharmacodynamics research.
