摘 要:目的观察红花对肝星状细胞c-fos和c-jun基因表达的影响。方法在培养的肝星状细胞株中分别加入不同浓度的红花(终质量浓度分别为1.0、0.5g/L);于8、24、48、72h4个时间点分别收集细胞,提取肝星状细胞总RNA;用逆转录.聚合酶链反应(RT-PCR)方法测定c-fos和c-jun的基因表达水平。结果红花两组HSC-T细胞c-fos基因表达水平在8h(0.49±0.13、0.34±0.11)、24h(0.42±0.10、0.26±0.08)、48h(O.31±0.07、0.20±0.04)、72h(0.23±0.05、0.12±0.04)4个时间点均明显低于对照组(0.63-t-O.13、0.50士0.12、0.43±0.06、0.32±0.04);c-jun基因表达水平在8h(0.70±0.14、0.46±0.11)、24h(0.57±0.10、0.43±0.07)、48h(0.42±0.09、0.26±0.06)、72h(0.31±0.05、0.12±0.04)4个时间点也均明显低于对照组(0.93±0.13、0.80-4-0.12、0.63±O.10、0.46±0.07);随剂量加大,HSC-T细胞c-fos、c-jun基因表达水平逐渐降低;两组间差异有统计学意义(P〈0.05)。结论红花对肝星状细胞c-fos和c-jun基因表达有明显下调作用。Objective To investigate the effects of carthamus tinctorius on c-fos and c-jun gene expression in hepatic stellate cells (HSCs). Methods Hepatic stellate cell' line HSC-T6 cells were cultured in medium containing different concentrations of carthamus tinctorius ( 1.0, and 0. 5 g/L) and harvested at differeni incubation periods (8, 24, 48, and 72 h). Total RNA of HSCs was isolated, and c-fos and c-jun gene expression levels were detected by using reverse transcription polymerase chain reaction. Results C-fos gene expression levels of HSCs in two carthamus tinctorius-treated groups were significantly lower than in control group (0. 63 ± 0. 13, 0. 50 ± 0. 12, 0.43 ± 0.06, 0. 32 ± 0. 04 ) after exposure to carthamus tinctorius for 8 h (0.49 ± 0. 13, 0. 34 ± 0. 11 ), 24 h (0.42 ± 0. 10, 0. 26 ± 0.08), 48 h (0. 31 ± 0. 07, 0. 20 ± 0.04) , and 72 h (0. 23 ± 0.05, 0. 12 ± 0. 04 ). C-jun gene expression levels of HSCs in two carthamus tinctorius-treated groups were also significantly lower than in control group (0. 93 ± 0. 13,0. 80 ± 0. 12, 0. 63 ± 0. 10, 0. 46 ± 0.07 ) after exposure to carthamus tinctorius at 8 h ( 0. 70 ± 0-14,0.46±0.11), 24h (0.57±0.10, 0.43±0.07), 48h (0.42±0.09, 0.26±0.06), and 72h (0. 31 ±0. 05, 0. 12 ±0. 04). The gene express'ion levels of c-fos and c-jun in HSCs were decreased gradually as the dosage of carthamus tinctorius increased, and there were significant difference in the c-los, and c-jun gene expression between two carthamus tinctorius-treated groups. Conclusion Carthamus tinctorius may down-remalate c-fos and c-jun gene exoression in HSCs.
