摘 要:目的探讨流式细胞术研究白假丝酵母菌耐药机制的可行性。方法临床分离对氟康唑敏感的白假丝酵母菌种,经体外诱导产生耐药。应用荧光染料PI(碘化丙啶)和罗丹明123染色,通过流式细胞术检测敏感株、耐药株、回复敏感株的细胞周期和药物外排活性。结果与敏感株和回复敏感株比较,耐药株增殖活性明显下降,但是药物外排活性显著增强。结论应用氟康唑能够体外诱导白假丝酵母菌产生耐药。流式细胞术应用于白假丝酵母菌增殖活性和药物外排活性研究是可行的。Objective To observe the feasibility of research on the drug-resistance of Candida albicans by flocytometry.MethodsThe strain of fluconazol-sensitive Candida albicans was induced with fluconazol to get the drug-resistant filial generation in vitro.The drug efflux and the cell cycles of the sensitive strain,regained sensitive strain and the induced resistant strain,stained with PI and rhodamine 1,2 and 3,were detected by flowcytometry.Results Compared with the sensitive strain and regained sensitive strain,the proliferation activity of the resistant strain was decreased but the drug efflux was increased.Conclusion Susceptible Candida albicans can develop fluconazole resistance when exposed to fluconazole in vitro.Flowcytometry can be used to study the activities of the drug efflux and proliferation in Candida albicans.
