摘  要:目的:探讨TLR4的激活对哮喘血清被动致敏人气道平滑肌细胞(HASMCs)增殖与合成分泌功能的影响。方法:通过培养原代HASMCs,以人哮喘血清致敏细胞,观察HASMCs表面TLR4的表达、细胞增殖反应与转化生子因子-β1(TGF-β1)在哮喘状态下的合成分泌水平。结果:与正常对照组相比,被动致敏组和TNF-α组HASMCsTLR4表达均显著增多(P<0.01),增殖反应显著增强(P<0.01),总蛋白浓度、IgE与TGF-β1的分泌水平均显著增高(P<0.01),并且TNF-α组各项指标均较被动致敏组效果更加显著;抗TLR4抗体组各项指标较被动致敏组、TNF-α组显著减弱或减少(P<0.01)。结论:哮喘血清被动致敏HASMCs表面TLR4的活化可能通过诱导细胞的增殖和TGF-β1的合成分泌,导致哮喘气道微环境的改变,参与哮喘气道重构过程。Objective: To investigate the activation of Toll like receptor 4(TLR4) on passively sensitized human airway smooth muscle cells(HASMCs) proliferation and the synthesis and secretion function.Methods: Through the cultivation of primary HASMCs,we studied TLR4 expression on cell surface,cell proliferation and transformation of parturient factor-β1(TGF-β1) in asthma under the condition of synthesis and secretion level by passively sensitized HASMCs with asthma serum.Results: Compared with the control group,in passive sensitized group and TNF-α group TLR4 expression were significantly increased(P0.01),significantly enhanced proliferation(P0.01),total protein concentration,IgE secretion and TGF-β1 were significantly higher(P0.01);and all the above parameters were increased more significantly in TNF group compared with those in the target effect of passively group;and those parameters were significantly reduced in anti-TLR4 antibody group compared with those in the target effect both of passively sensitized group and TNF-α group.Conclusion: TLR4 on passively sensitized HASMCs activated can induce the excessive proliferation of HASMCs and a large number of synthesis and secretion of TGF-β1,resulting in changing airway micro-environ-ment,which involved in airway remodeling in asthma.
