摘  要:目的:探讨补肾中药合剂对体外培养的人成骨细胞(hFOB 1.19)功能影响的分子机制。方法:分别对低剂量、高剂量补肾中药血清和5%空白对照组血清培养的成骨细胞进行碱性磷酸酶(ALP)测试,并通过酶联免疫吸附实验分别检测其成骨细胞骨钙素(OC)的分泌量。采用蛋白质印迹法分析补肾中药合剂含药血清对骨保护素(OPG)、骨桥蛋白(OPN)和核因子κB受体活化因子配体(RANKL)蛋白表达水平的影响。结果:补肾中药合剂含药血清能提高hFOB 1.19细胞中的ALP活性;作用96 h后,能促进hFOB 1.19细胞分泌OC,均具有剂量依赖关系;在蛋白表达水平上,补肾中药合剂含药血清能明显上调人成骨细胞的OPG、OPN,并抑制其RANKL的表达。结论:补肾中药合剂含药血清可增张成骨细胞的成骨功能,其机制可能与影响OPG/RANK/RANKL骨代谢信号调节系统有关。Objective: To study the effects of the medicated serum of a kidney-reinforcing chinese herbal mixture on the function of human osteoblastic cells hFOB 1.19 in vitro and to explore the relevant molecular mechanisms. Methods: Medicated serum was pre- pared using different doses of a kidney-reinforcing chinese herbal mixture in SD rats. hFOB 1.19 cells were treated by the medicated sera. Cellular alkaline phosphatase (ALP) activity, Osteocalcin(OC) secretion and the protein levels of osteoprotegerin (OPG) , os- teopontin (OPN) , and receptor activator of NF-KB ligand (RANKL) were examined by a ALP kit, ELISA and Western blot respec- tively. Results: The medicated serum enhanced ALP activity in hFOB 1.19 cells at the low dose (P 〈0.05) and high dose (P 〈 0.01 ). Exposing hFOB 1.19 cells to the medicated sera for 48 h did not result in any statistical difference in OC secretion among the control and experimental groups; however, a 96 h exposure to the medicated sera promoted OC secretion( compared with the mock con- trol, low-dose serum, P 〈0.05, and high-dose serum, P 〈0.01 ). The medicated sera increased the expressions of OPG and OPN ( P 〈 0.05 ) , but suppressed RANKL expression(P 〈 0. 01 ) in hFOB 1.19 cells. Conclusion : The medicated sera of the kidney- re- inforcing chinese herbal mixture may promote the differentiation and function of osteoblasts.The underlying molecular mechanism is associated with singnal regulating system of OPG/RANK/RANKL.
