摘 要:研究Th17型细胞因子白细胞介素-8(IL-8)、IL-17和IL-23受体(IL-23R),探讨其在非嗜酸细胞性哮喘发病机制中的作用。搜集26例非嗜酸细胞性哮喘患者、28例嗜酸细胞性哮喘患者和28例健康者的诱导痰及外周静脉血,离心后取痰上清液及血清,采用酶联免疫吸附实验(ELISA)法测定诱导痰上清液及血清中IL-8、IL-17和IL-23R的浓度。HE染色法观察气道粘膜上皮的炎症浸润情况。与健康者相比,非嗜酸细胞性哮喘病人气道组织病理学检查示大量炎症细胞浸润,其血清和诱导痰上清液IL-8、IL-17和IL-23R水平均明显增高(P<0.05),而嗜酸细胞性哮喘患者血清和诱导痰上清液IL-8、IL-17和IL-23R水平无显著变化(P>0.05)。Th17型炎症因子IL-8、IL-17和IL-23R在非嗜酸细胞性哮喘发病机制中具有重要作用。To evaluate the expression of Thl7-related cytokines such as interleukin-8(IL-8), IL-17 and IL-23 receptor(IL-23R) in asthmatic patients and investigate the effects and underlying mechanisms in non-eosinophilic asthma (NEA), the serum after centrifuging peripheral blood and induced sputum from 26 non-eosinophilic asthma patients, 28 eosinophilic asthma patients and 28 healthy control persons were collected. The concentration of IL-8, IL-17 and IL-23R was measured respectively by enzyme- linked immunosorbent assay (ELISA). The airway inflammation was observed with hematoxylin and eosin (HE). Compared with healthy subjects, the leve!~ of IL-8, IL-17 and IL-23R in the serum and induced sputum of non-eosinophilic asthma patients were significantly increased (P〈0.05), while the eosinophilic asthma patients showed no obvious changes (P〉0.05). We also observed more inflammatory cells stained with HE in airway epithelium of asthmatic patients compared with healthy control subjects (P〈0. 001). Thl7-related cytokines such as IL-8, IL-17 and IL-23R might play pivotal roles in bronchial non-eosinophilic asthma mechanism.
