摘 要:为了提高解淀粉芽孢杆菌Bacillus amyloliquefaciens BW-13抗真菌活性物质的产量,本实验首先用单因素研究不同碳源,氮源和微量元素对BW-13产生抗真菌活性物质的影响,并使用均匀设计法对培养基进行优化.结果表明:玉米粉和低浓度的Mn2+对BW-13产生抗真菌活性物质的促进效果明显.最佳培养基配方和发酵条件为:玉米粉32g/L,蛋白胨9g/L,氯化锰5mg/L,初始发酵pH 5.0,装液量30mL,接种量为4%,培养温度28℃,摇床转速200r/min,培养时间144h.优化后5μL BW-13发酵液抑菌圈提高到28.5mm,比优化前提高了28.94%.Object: To optimize medium and fermentation conditions for the production of antifungal substance produced by Bacillus amyloliquefaciens BW-13, single factor experiments including different carbon sources, nitrogen sources and metal ions were tested. When corn starch and Mn^2+ were used, an obviously increasing of antifungal substance was observed. The medium and fermentation conditions optimized with uniform design were as follows: corn starch 32 g/L, peptone 9 g/L, manganese chloride 5 mg/L, initial pH 5.0, liquid voiume 30mL in 250 mL flask, inoculum size 4%, incubation temperature 28 ℃, shaking speed 200 r/min, culture time 144 h. After optimization, the inhibition zoom diameter by 5μL of BW-13 fermentation broth was reached to 28.5 mm,increased by 28.94% compared to the control.
