摘 要:目的建立小鼠下腔静脉深静脉血栓(DVT)模型,观察血栓形成后下腔静脉及血栓栓子中巨噬细胞炎性蛋白-1α(MIP-1α)的表达及其与血栓栓子溶解的关系。方法 100只18~22g的昆明种小白鼠(雌雄不限)随机分为对照组(n=10)和模型组(n=90),模型组采用神经血管钳钳夹联合结扎下腔静脉建立小鼠DVT模型。建模后将模型组分为DVT组(小鼠不加任何干预,n=45)和DVT+MIP-1α-Ab组(小鼠予以MIP-1α中和抗体持续腹腔注射7d,n=45),于2、4、8、12、21d5个时间点各处死8只,质量/长度比值比较栓子溶解程度,应用SYBR?GreenⅠ核酸凝胶染液实时荧光定量PCR及ELISA检测股静脉组织中MIP-1αmRNA、蛋白的表达量。结果 DVT组1只小鼠因严重感染死亡;模型组小鼠均有血栓形成。DVT组及DVT+MIP-1α-Ab组栓子质量/长度比值随时间的延长呈逐渐缩小的趋势(P<0.05),相同时间点DVT组血栓栓子质量/长度比值减小更为明显(P<0.05);DVT组MIP-1αmRNA、蛋白随时间点的改变呈逐渐升高趋势,8d升至最高点(P<0.05),后呈逐渐下降趋势,DVT+MIP-1α-Ab组MIP-1αmRNA、蛋白表达亦呈上述趋势,但差异无统计学意义。相同时间点组间比较,DVT组MIP-1αmR-NA、蛋白变化较为明显(P<0.05)。结论 DVT形成后MIP-1α参与了血栓栓子的溶解过程,MIP-1α中和抗体可阻断其溶栓效应。Objective This study was to establish mice deep vein thrombosis model,and after thrombosis,to observe the expression of MIP-laof inferior vena cava and thromboembolus, and reveal the relation between the expression and the dissolution of thromboembolus. Methods 100 mice whose weight rage between 18--22 g were randomly divided into normal control group (n= 10) and model group (n=90). The mice of the model group were established deep vein thrombosis model by clamping with the neurovascular clamp combined with ligation in the inferior vena cava. Then the model group was divided into 2 subgroups: DVT group (mice without any intervention, n = 45) and DVT+ MIP-1 a-Ab group (mice received MIP-1 a antibody intraperitoneal injection for 7 d, n = 45), executed 8 mice on the 2 d, 4 d, 8 d, 12 d, and 21 d time points respectively, and quality/length ratio were used to compare the embolus dissolve degree. The SYBRa Green I fluorescence real-time quantitative PCR and ELISA were used to detect the expression levels of MIP-lamRNA and protein in the femoral vein organization. Results There was one mouse in the model group dead because of serious infection, and there was thrombosis in every mouse of model group. The embolus quality/length ratio of DVT group and DVT+MIP-la-Ab group showed a diminishing trend along with the extension of time(P〈0.05) ,and at the same time point the decrease of the thromboembolus quality/length ratio of DVT group was more obvious (P〈0.05). As the time point changed, the expression of MIP-1 a mRNA and protein of DVT group showed an increasing trend, which reached the highest point in the 8th day(P〈0.05) and then declined gradually. The expression of MIP-1 a mRNA and protein in DVT + MIP-la-Ab group also showed an increasing trend while the difference had no statistical significance. The changes of MIP-1 a mRNA and protein of DVT group were more obvious compared with DVT+MIP la-Ab group at the time point(P〈0.05). Conclusion The results demonstrated that
