摘 要:目的研究中药参仙合剂对环磷酰胺模型小鼠骨髓抑制和血小板生成素的影响。方法将昆明种小鼠随机分为六组,每组8只,即空白组、模型组、阳性对照组、参仙合剂低剂量、中剂量、高剂量组。参仙合剂各组先予灌胃给药5 d,第6天开始除空白组外各组均予环磷酰胺100 mg.kg-1腹腔注射,连续3 d造模,阳性对照组皮下注射重组白细胞介素-11(rh LI-11)200μg.kg-1,连续6 d,参仙合剂各组继续灌胃给药6 d。定期检测各组小鼠白细胞(WBC)和血小板计数(PLT),并于实验第12天检测小鼠骨髓巨核细胞数和血清及骨髓上清液中血小板生成素(TPO)。结果参仙合剂各组WBC、PLT和巨核细胞数显著高于模型组(P<0.05或P<0.01);阳性对照组巨核细胞数显著高于模型组(P<0.01)。参仙合剂各组与阳性对照组血清及骨髓上清中TPO浓度显著高于模型组(P<0.01),TPO与参仙合剂剂量正相关,具有显著的量效关系。结论参仙合剂可提高骨髓与血液中TPO水平,从而促进巨核细胞和血小板等生成,防治骨髓抑制。Objective To study the impacts of shenxian mixture on the bone marrow suppression and thrombopoietin(TPO)in cyclophosphamide induced model mice.Methods Kunming mice were randomized into six groups,8 mice in each one,named a blank group,a model group,a positive control group,a low-dose group,a middle-dose group and a high-dose group of shenxian mixture.In each shenxian mixture group,the drug lavage was done for 5 days.Since the 6th day,the intraperitoneal injection of cyclophosphamide 100 mg·kg-1 was applied in each group except the blank group,continuously for 3 days for modeling.In the positive control group,the subcutaneous injection of recombinant interleukin-11(rh LI-11),200 μg·kg-1 was given,continuously for 6 days.The drug lavage was continued in each shenxian mixture group for the other 6 days.The white blood cell(WBC)and platelet(PLT)counts were detected regularly for the mice in each group.On the 12th day of the experiment,the bone marrow megakaryocyte count and TPO concentration in the serum and bone marrow supernatant were detected.Results WBC,PLT and megakaryocyte counts in each shenxian mixture group were higher significantly than those in the model group(P0.05 or P〈0.01).The megakaryocyte count in the positive control group was higher significantly than that in the model group(P〈0.01).TPO concentration in the serum and bone marrow supernatant in each shenxian mixture group and the positive control group was higher significantly than that in the model group(P〈0.01).The positive relationship was presented between TPO concentration and the dose of shenxian mixture,indicating the significant dose-effect relationship.Conclusion Shenxian mixture improves TPO level in bone marrow and blood so that it promotes the formation of megakaryocytes and platelets,prevents and treats bone marrow suppression.
