摘 要:目的:测定氧化槐果碱在正辛醇-水和正辛醇-缓冲液体系中的表观油水分配系数,为体内吸收研究提供参考。方法:以HPLC-DAD为检测手段,采用摇瓶法测定氧化槐果碱的表观油水分配系数。结果:在(37±1)°C下,氧化槐果碱在正辛醇-水体系中的表观油水分配系数为logPapp=-0.44。在pH 1.2~10.0的磷酸盐缓冲液中,测得logPapp在-0.97~-0.25;当pH<4.5时,受pH影响不显著,表现为亲水性;当pH>4.5时,logPapp随pH增大而逐渐增大。结论:该法可准确、快速测定氧化槐果碱的表观油水分配系数,并借此解析其体内动态。Objective: To determine the apparent oil-water partition coefficient of oxysophocarpine in n- octanol-water and n-octanol-phosphate buffer system, and to provide reference for the absorption study in vivo. Method: The apparent oil-water partition coefficient for oxysophocarpine was performed by using shaking flask method with HPLC-DAD as separation and detection pathway. Result: The apparent oil-water partition coefficient of oxysophocarpine was log Papp = - 0.44 in n-octanol-water system at 37 ℃. In pH 1.2-10.0 phosphate buffer solutions, log Papp was in the range of - 0.97- - 0.25. When pH 〈 4.5, there was no significant effect on it by pH. When pH 〉4.5, log Papp of oxysophocarpine showed the gradual increasing tendency with the increasing of pH. Conclusion: The methods described in the paper can be used to determine the oil-water partition coefficient of oxysophocarpine accurately, and help to predict the intracoporal process.
