摘 要:目的:探讨从具有抗类风湿关节炎作用的宽叶荨麻中分离到的9个化学成分对人类风湿性关节炎滑膜成纤维细胞MH7A增殖的影响。方法:分别以从宽叶荨麻中分离得到的9个化学成分(0~400 mg.L-1):RoseosideⅡ(C1),(6S,9R)-3-氧-α-紫罗兰醇-β-D-葡萄糖苷(C2),(6R,9R)-3-氧-α-紫罗兰醇-β-D-葡萄糖基(1→2)-β-D-葡萄糖苷(C3),芹菜素-6,8-二-C-β-D-吡喃葡萄糖苷(C4),木樨草素-7-O-新橙皮糖苷(C5),木樨草素-7-O-β-D-吡喃葡萄糖苷(C6),芦丁(C7),异落叶松脂素-9-O-β-D-吡喃葡萄糖苷(C8),5α,6β-二羟基-胡萝卜苷(C9)处理MH7A细胞(2.5×104/mL),24 h后采用MTT法检测细胞增殖率,并通过测定DNA片段化检测细胞凋亡率。结果:在被检测的9个化合物中,化合物5α,6β-二羟基-胡萝卜苷显示了较强的抑制MH7A细胞增殖的作用,其IC50值为140 mg.L-1。DNA片段化分析结果表明,其能显著诱导MH7A细胞凋亡。结论:宽叶荨麻中化学成分5α,6β-二羟基-胡萝卜苷能够显著抑制MH7A细胞增殖并能诱导MH7A细胞凋亡。Objective: The antiproliferative and apoptosis inducing effects of the compounds isolated from Urtica laetevirens on MH7A human rheumatoid arthritis synovial cells were tested in this study. Method: MH7A cells were treated with nine compounds isolated from U. laetevirens, respectively. The antiproliferative activities of nine compounds were evaluated by MTT assay, and the apoptosis was assessed with DNA fragmentation after 24 h. Result : Among the nine compounds 5a, 6fl-dihydroxy-daucosterol showed stronger antiproliferative activity against MH7A ceils. It could induce DNA fragmentation in MH7A cells. Conclusion: The compound, 5a, 6/3-dihydroxy- daucosterol isolated from U. laetevirens could inhibit cell proliferation and induce apoptosis in MH7A cell.
