摘 要:目的观察葡萄籽原花青素(Grapeseed proanthocyanidins extracts,GSPE)对2型糖尿病大鼠肾脏的影响,并进一步探讨其可能的作用机制。方法雄性SD大鼠高脂饲料喂养4周后,腹腔注射链脲佐菌素(STZ,0.01mol/L,30mg/kg.bw)建立糖尿病大鼠模型,成模后随机分为糖尿病对照组(DM对照组)、GSPE低、中、高剂量干预组,基础饲料饲养的大鼠为正常对照组。GSPE低、中、高剂量组的剂量分别为125、250、500mg/kg.bw。喂养24周后,测定各组大鼠的肾功能指标,肾组织的抗氧化指标及血清超敏C反应蛋白(hs-CRP)含量。结果 GSPE各组大鼠空腹血糖值、24h尿蛋白、血清尿素氮、血肌酐和右肾指数低于DM对照组(P﹤0.05),尿肌酐含量明显高于DM对照组;GSPE低剂量组大鼠的肾脏SOD活性明显高于DM对照组(P﹤0.01),GSPE各剂量组大鼠肾脏MDA含量明显低于DM对照组(P﹤0.01),GSPE低剂量和高剂量组大鼠肾脏CAT含量高于DM对照组(P﹤0.05);GSPE中剂量和高剂量组大鼠血清hs-CRP含量显著低于DM对照组(P﹤0.01)。结论 GSPE对实验大鼠2型糖尿病肾病有一定的改善作用,并可有效地降低其炎症水平。OBJECTIVE To study the effects of grape seed proanthocyanidins extracts (GSPE) on experimental type 2 diabetic nephropathy in rats and its possible mechanism. METHODS To establish type 2 diabetic rats model, male SD rats were fed on high fat diet for 4 weeks, and were injected with streptozotocin (STZ, 0.01mol/L, 30mg/kg·bw). Ninety-six diabetic rats were randomly divided into diabetes-vehicle group (DM control group), three GSPE-treated groups. Twenty-four normal rats were enrolled as the normal control group. The doses in three GSPE-treated groups are 125, 250, 500mg/kg.bw. After 24 weeks, renal tissue antioxidant indexes and serum high sensitive C reactive protein (hs-CRP) content were measured, and the renal function indexes were tested as well. RESULTS Compared with the DM control group, the fasting blood glucose values, the urinary protein/24h, levels of blood urea nitrogen (BUN), serum ereatinine (SCr) and the ratio of fight kidney weight/ body weight decreased (P 〈 0.05), urinary creatinine (UCr) was significantly raised in threeGSPE-treated groups (P 〈 0.01 ), the SOD activity in kidney was raised (the GSPE low dose group, P 〈 0.01 ), CAT content also increased (the GSPE low and high dose groups) while MDA content decreased in the GSPE group (P 〈 0.01 ), and the differences were all significant. The serum hs-CRP level in GSPE groups were significantly lower than that in normal control group and DM control group (P 〈 0.01 ). CONCLUSION GSPE has improvement effect on experimental rats with type 2 diabetic nephropathy, and can effectively re- duce inflammatory reaction.
