摘  要:目的观察薯蓣皂苷片对胶原诱导性关节炎(collagen-induced arthritis,CIA)大鼠滑膜组织血管内皮细胞生长因子(vascular endothelial growth factor,VEGF)表达的影响,探讨薯蓣皂苷片对类风湿关节炎抗血管新生的机制。方法 48只Wistar大鼠随机分成4组:正常对照组(A组),模型组(B组),阳性药物对照组(C组),薯蓣皂苷片组(D组),每组12只。B、C、D组大鼠均建立CIA大鼠模型,待模型建立成功后,C、D组大鼠分别给予相应的药物灌胃,连续21d,采用SP免疫组化方法通过CD31给滑膜微血管密度(MVD)计数,并采用实时荧光定量PCR方法检测关节滑膜组织VEGF的表达。结果 B组大鼠滑膜组织MVD、VEGF显著高于A组(P﹤0.01);C、D组大鼠滑膜组织MVD、VEGF明显低于模型组(P﹤0.01);C、D组之间差异无统计学意义(P﹥0.05)。结论薯蓣皂苷片可以通过降低大鼠滑膜组织VEGF的表达发挥抑制滑膜血管新生的作用。OBJECTIVE To study effects of Dioscornin Tablets on expression of vascular endothelial growth factor (VEGF) in synovial tissue of collagen-induced arthritis (CIA) rats, and to investigate the mechanism of Dioscornin Tablets antiangiogene- sis in treating rheumatoid arthritis. METI-IEDS 48 wistar rats were randomly divided into 4 groups (n = 12/group) : normal control group (A), CIA model group (B), tripterygium group (C), Dioscornin Tablets (D). Rats in B-D group were estab- lished CIA rat model and the rats in C-D group were lavaged with corresponding medicine for 21 days. The micro-vessel density (MVD) of synovium was measured by caulating CD31 level and real-time PCR was used to detect VEGF expression in synovial tissue of CIA rats. RESULTS MVD, VEGF expression in synovial tissue of CIA rats in B group were signficantly higher than A group (P 〈 0.01) ; MVD, VEGF expression in synovial tissue of CIA rats in C-D group were significantly lower than B group (P 〈 0.01). Moreover, there were no significantly differences between C group and D group (P 〉 0.05). CONCLUSION Dioscornin Tablets can inhibit angiogenesis in synovial tissue by down regulating VEGF expression.
