系膜细胞膜固相色谱法研究丹皮中的效应成分

摘　　要：丹皮常被用于治疗肾病的中药复方中,但其活性成分并不明确,因此,采用肾小球系膜细胞膜固相色谱法分析丹皮中的效应成分.培养小鼠肾小球系膜细胞,并作为固定相选择性结合丹皮中的活性成分,排除未与细胞膜靶点结合的非活性成分后,使细胞膜靶点失活,解离系膜细胞膜上被结合的活性成分,用UPLC对解离液进行分析.结果表明丹皮酚是丹皮与肾系膜细胞结合的成分,因此认为丹皮酚是丹皮中的效应成分.Moutan Cortex is a herb commonly used in the prescriptions on treating nephropathy.In order to analyze the bioactive compounds in Moutan Cortex,mouse renal mesangial cells were cultured and used to bind and separate components in Moutan Cortex extraction.The cells were washed to take away the unbounded components,then the dissociative compounds from mesangial cells were analyzed by UPLC.One compound which could interact with mesangial cells was found,and it was identified as paeonol.Therefore,we conclude that paeonol is the major bioactive compound in Moutan Cortex.
