摘 要:利用前期构建的SSR核心引物的指纹图谱为基础,从中选出123份具有代表性的水稻品种进行品种的特异性及稳定性分析,并据此判定新增核心标记的应用价值。聚类分析结果表明,24对引物共检测到138个等位基因,其中等位位点上只存在一个品种有12个,能特定区分粳稻和籼稻类群水稻品种的等位位点的有21个;按遗传相似系数为0.96的划分标准,所有的品种都具备特异性;以等位位点的纯合性为指标,117份品种具有稳定的遗传基础;在杂交种与其父母本的遗传位点匹配试验中,16份随机配组的杂交组合中含有1~3个不匹配的遗传位点,而且这些位点和6份品种的杂合位点一起,多集中于RM289、RM7492、RM6487和RM583等4个标记中,它们是品种遗传不稳定的分子基础。表明24对核心引物能较好的鉴别123份水稻品种,123份品种中含有可重复发生的不稳定遗传位点。The 123 representatives of rice varieties were chosen to conduct the analysis of cultivar specificity and stability based on the fingerprinting profiles built previously by SSR core primers, in order to determine the value of the new adding core markers. The cluster analysis showed that 24 pairs of primers detected 138 alleles, of which there are 12 loci happening in single cultivar and 21 loci that can distinguish japonica and indica rice varieties. Based on criterion of genetic similarity coefficient as 0.96 for the classification, all tested varieties possess their specificity; based on homozygosity of alleles as reference, 117 varieties have stable genetic basis. In the experiment of match analysis on hybrid and their parents, 16 hybrids by random combining contain 1 and or 3 unmatch genetic loci that are happening with heterozygous loci of six varieties, which mostly appear in the markers of RM289, RM7492, RM6487 and RM583. We thought that these are the molecular basis of genetic instability of the varieties indicating 24 pairs of core primers should well identify 123 varieties of rice, and also 123 varieties might contain repeatable instable genetic loci.
