摘 要:目的探讨六味地黄生物制剂延缓衰老的机制。方法小鼠随机分为正常对照组、阳性药物(维生素E)组、衰老模型组与六味地黄生物制剂组。除正常对照组注射生理盐水外,其余各组皮下注射0.5mL 5%D-半乳糖以建立衰老模型,连续50 d。第10天开始以0.2 mL.10 g-1给予各组小鼠灌胃相应的药物,50 d后处死小鼠,分离脾淋巴细胞并利用单细胞凝胶电泳法检测各组小鼠脾脏淋巴细胞的DNA损伤情况。结果与正常对照组比较,衰老模型组小鼠的脾淋巴细胞有较为严重的损伤(P<0.01),尾长、尾距与DNA拖尾率分别为(145.96±94.63)μm、(51.34±44.67)μm、(23.14±15.90)%;与衰老模型组比较,阳性药物组和六味生物制剂组小鼠的DNA损伤均显著降低(P<0.01)。结论六味地黄生物制剂对D-半乳糖所致的衰老小鼠的脾淋巴细胞有明显的保护作用,这可能是六味地黄生物制剂延缓衰老的机制之一。Objective To study the mechanism of LW-PSB in delaying senescence.Method Mice were randomly divided into 4 groups,including the control group,the positive drug group,the aging model group and the LW-PSB group.Animal model was established by subcutaneous injection with 0.5 mL 5% D-gal once daily for 50 consecutive days,except for the control group which was treated with saline.Mice in each group began to be given corresponding drug at 0.2 mL·10 g^-1 dosage at day 10.Spleen lymphocytes were extracted and subjected to electrophoresis to analyzing the DNA damage at day 50.Results Compared with the control group,spleen lymphocytes in aging model of mice were serious damage(P〈0.01),and the tail length,tail moment and tail DNA% were(145.96±94.63) μm,(51.34±44.67) μm and(23.14±15.90)%,respectively.Compared with the model group,changes of these parameters in the positive drug group and the LW-PSB group were significant(P〈0.01).Conclusion LW-PSB can obviously protect against DNA damage of spleen lymphocytes in aging model mice induced by D-galactose that may be one of the molecular mechanisms by which LW-PSB delays senescence.
