摘  要:目的:研究还原型谷胱甘肽对大鼠急性肺损伤(acute lung injury,ALI)的保护作用。方法:SD大鼠24只,随机分为假手术组、模型对照组和研究组。模型对照组和研究组制作失血性休克模型,造成ALI,然后在20 min内复苏,研究组复苏时应用还原型谷胱甘肽;假手术组只进行相应的手术操作。测定各组大鼠动脉血浆中肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)含量、支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF)中巨噬细胞(pulmonary alveolar macrophages,PAM)和中性粒细胞(polymorphonuclear leukocyte,PMN)总数、BALF中蛋白含量和肺组织中超氧化物歧化酶(superoxide dismutase,SOD)含量;计算肺组织含水率;光镜下观察肺组织形态学改变。结果:与假手术组比较,复苏后模型对照组、研究组血浆中TNF-α含量、BALF中的蛋白含量和PMN总数、肺组织含水率显著升高,SOD含量显著降低,但研究组的变化程度均显著小于模型对照组。光镜下可见,模型对照组肺组织间质水肿、细胞浸润明显,而研究组肺组织的上述改变较轻。结论:还原型谷胱甘肽对SD大鼠的急性肺损伤具有保护作用。Objective:To study the protective effects of reduced glutathione on acute lung injury(Aid) in rats. Methods:Twen- ty -four SD rats were divided into 3 groups randomly. One group was non-operation group. Hemorrhagic shock models were performed in both model control group and study group. Then the shock rats were resuscitated in 20 rain. Reduced glutathione was used in study group during resuscitation. The arterial blood of rats was taken in different time to determine the plasma tumor necrosis factor-α(TNF-α) concentration. The amount of pulmonary alveolar macrophages (PAM) and polymorphonucle ar leukocytes (PMN) in bronchoalveolar lavage fluid (BALF) were counted at the end of experiment . At the same time , the protein contents in BALF and superoxide dismutase(SOD) contents in lung tissues were measured , and the water ratio of lung tissues was calculated. The histological morphology of lung tissues were examined under optical microscope. Results: Corn: pared with non-operation group,in study group and model control group,TNF-α concentration , the protein contents and the a- mount of PMN in BALF, and the water ratio in lung tissues went up obviously after resuscitation SOD contents of lung tissues went down, more significantly in model control group. Lung tissue edema and neutrophil accumulation have be observed both in model control group and study group, but the changes in the latter were less serious. Conclusions: Reduced glutathione has protective effects on ALI in SD rats.
