绵羊红细胞溶血试验用于化妆品眼刺激试验的动物替代方法研究

摘　　要：采用绵羊红细胞(SRBC)体外溶血试验测定了32种不同品类化妆品筛选配方或终配方样品的半数溶血浓度(HC5)0,并与家兔眼刺激试验结果比较,研究了SRBC体外溶血试验替代动物眼刺激试验的可靠性,初步确定其判断标准。32个样品中有30个样品可测得其HC50,1个样品中含有红色物质,另有1个样品不能导致血红蛋白变性而不能测得其HC50。家兔眼刺激试验结果表明,32个样品中有5个样品具有轻微眼刺激性,其余样品均无眼刺激性。该5个样品的HC50在0.017%～0.026%之间,-logHC50≥3.59,其余25个无眼刺激性样品的HC50>0.026%,-logHC50<3.59。结果显示,SRBC溶血试验可以用于替代化妆品的动物眼刺激试验。This paper studied the reliability of sheep blood cells （SRBC） haemolysis test, as an in vitro alternative method, to replace the eye irritation method-in vivo Draize rabbit test, for the evaluation of cosmetic products, and set the criterion for sheep blood cells haemolysis test. The 50% of haemolytic concentrations （HC50） of 32 cosmetic samples supplied by the product development department were tested and the results were compared with that of Draize rabbit test. The results showed that 30 HC50 values of 32 samples have been determined accurately according to Hygienic Standard for Cosmetics （2007）; the other two alues cannot be determined because one sample contained reddish substance in it and another cannot induce haemoglobin denaturation. Draize rabbit test indicated that 5 samples were mild in eye irritation and their HC50 were between 0.017% - 0.026%（W/V）, -logHCs0 were equal to or higher than 3.59. The other 25 samples were classified as no eye irritation and their HC50 were higher than 0.026%（W/V）, -logHCso were lower than 3.59. Conclusion showed that sheep red blood cell haemolysis test can be used as an alternative method for evaluation of eye irritation of a cosmetic product.
